银木种子园及基于SNP标记的遗传分析

摘  要:银木是我国南方珍贵用材和特用经济树种,作为国家级良种基地的重庆市大足区西山林场建立了中国第一个银木种子园,建园亲本来自重庆大足区、铜梁区、宜宾市、高县、江安县、宜宾县、兴文县等地的优树材料。为了弄清这些个体间的亲缘关系及群体的遗传多样性水平,本研究以银木同属物种沉水樟的基因组作为参考基因组,采用GBS测序策略开发了银木SNP分子标记。结果表明:银木与沉水樟基因组平均比对率为96.13%。经条件过滤,我们共获得127825个SNP位点,利用这些位点信息构建了59个亲本的亲缘关系图谱,并进行了群体杂合度和核苷酸多样性分析。亲本群体期望杂合度(He)和观测杂合度(Ho)分别为0.1696和0.1916;核苷酸多样性(pi)为0.1712。研究结果为今后银木种子园改良和良种选育提供了基础数据和信息。Cinnamomum septentrionale is a valuable timber and special economic tree species in South China.As a national improved variety of forest tree,the first seed orchard of Cinnamomum septentrionale in China has been established in Xishan Forest Farm of Dazu District of Chongqing City.The parents of the seed orchard are excellent tree materials from Dazu District,Tongliang District,Yibin City,Gaoxian County,Jiang an County,Yibin County,and Xingwen County,etc.In order to clarify the genetic relationship between these individuals and the level of genetic diversity of the population,this study took the genome of Cinnamomum micranthum as the reference genome,and adopted the GBS sequencing strategy to develop the SNP molecular marker of Cinnamomum septentrionale.The results showed that the average ratio of genome comparison between Cinnamomum septentrionale and Cinnamomum micranthum was 96.13%.127825 SNP sites in total was obtained after conditional filtering.Using these SNPs information,this study constructed a genetic relationship map of 59 parents,and further carried out population heterozygosity and nucleotide diversity analysis.The expected heterozygosity(He)and observed heterozygosity(Ho)of the parent population were 0.1696 and 0.1916,respectively.The nucleotide diversity(pi)was 0.1712.Those results provided basic data and information for the improvement of seed orchard and the breeding of improved varieties of Cinnamomum septentrionale in the future.
