复方幺川调免消瘰颗粒的制备与鉴别及促小鼠脾淋巴细胞体外增殖的研究

摘　　要：目的筛选复方幺川调免消瘰颗粒的制备工艺,获得薄层色谱鉴别方法,以及对小鼠脾淋巴细胞体外增殖的影响。方法采用水提工艺获得复方幺川调免消瘰提取物冻干粉,通过正交实验法优化颗粒剂的处方;利用薄层色谱鉴别方法鉴别复方颗粒;用CCK-8法检测复方幺川调免消瘰颗粒对小鼠脾淋巴细胞体外增殖的影响。结果正交实验结果显示,辅料可溶性淀粉与提取粉末用量比为1∶1,使用70%的乙醇作为润湿剂,乙醇占冻干粉的重比为50%;获得复方中甘草与皂角刺的薄层色谱鉴别方法;复方幺川调免消瘰颗粒对LPS诱导下的小鼠脾淋巴细胞增殖具有促进作用。结论筛选工艺优化得到复方幺川调免消瘰颗粒,薄层色谱法鉴别出特征斑点,专属性良好,该复方对小鼠脾淋巴细胞体外增殖具有促进作用。Objective To explore the preparation technology of Compound Yaochuan Tiaomianxiaoluo Granules,develop an identification method using thin layer chromatography(TLC),and evaluate its effect on the proliferation of mouse spleen lymphocytes in vitro.Methods We utilized a water extraction process to obtain the lyophilized powder of Compound Yaochuan Tiaomianxiaoluo Granules.An orthogonal experiment method was employed to optimize the granule formulation.The identification of compound granules was carried out using the TLC identification method.The impact of the Compound Yaochuan Tiaomianxiaoluo Granules on the proliferation of mouse spleen lymphocytes in vitro was then assessed through CCK 8 method.Results The results of the orthogonal experiment indicated that the optimal ratio of excipient soluble starch to extract powder was 1∶1,while a wetting agent consisting of 70%ethanol and a weight ratio of ethanol to freeze-dried powder of 50%were also found to be most effective.Through TLC identification method of horny spines,we successfully identified Licorice and Gleditsiae Spina in the compound granules.Furthermore,our study confirmed that Compound Yaochuan Tiaomianxiao Granules can promote the proliferation of spleen lymphocytes.Conclusion The screening process was successfully optimized to obtain the Compound Yaochuan Tiaomianxiao Granules,and the characteristic spots were identified by TLC.Furthermore,our study demonstrated thatthe compound had a promoting effect on the proliferation of mouse spleen lymphocytes in vitro,thus,it provides reference for developing applications.
