摘 要:目的:通过网络药理学方法探讨大黄治疗糖尿病肾疾病(Diabetic Kidney Disease,DKD)的作用机制。方法:通过中药系统药理学数据库及分析平台(TCMSP)筛选大黄的活性成分及靶点;利用GeneCards和OMIM数据库检索DKD相关靶点;利用Cytoscape软件构建大黄治疗DKD的作用靶点与有效成分之间的关系网络;使用R软件进行基因本体论(GO)和京都基因与基因组百科全书(KEGG)富集分析大黄治疗DKD有效靶点的生物学功能和作用机制。结果:得到大黄有效成分8个,治疗DKD潜在靶点62个,DKD靶点11905个,大黄与DKD共同靶点59个。KEGG通路富集分析结果表明,大黄治疗DKD可能与肿瘤抑制蛋白p53信号通路和雌激素信号通路密切相关。结论:本研究初步验证了大黄治疗DKD的信号通路相关机制,为进一步研究提供了理论基础和科学依据。Objective:To explore the mechanism of Dahuang(Radix et Rhizoma Rhei)in the treatment of diabetic kidney disease(DKD)by means of network pharmacology.Methods:The active components and targets of Dahuang were screened through TCMSP,and the relevant targets of DKD were retrieved using GeneCards and OMIM databases.The relationship network between the effective targets and the effective components of Dahuang in treating DKD was constructed using Cytascape software,and the biological function and mechanism of the effective targets of Dahuang in treating DKD were analyzed by GO and KEGG enrichment using R software.Results:We obtained 8 active ingredients of rhubarb,with 62 potential targets for treatment of DKD,and 11905 targets for DKD,and 59 common targets for Dahuang and DKD.The enrichment results of KEGG pathway indicate that the treatment of DKD by Dahuang may be closely related to p53 signaling pathway and estrogen signaling pathway.Conclusion:This study preliminarily verified the signal pathway related mechanism of Dahuang in the treatment of DKD,and provided a theoretical basis and scientific basis for further research.
