基于网络药理学探讨柴胡疏肝散治疗乙型肝炎相关性肝癌的机制

摘　　要：目的:运用网络药理学方法探讨柴胡疏肝散治疗乙型肝炎相关性肝癌的作用机制。方法:通过TCMSP平台筛选柴胡疏肝散活性成分及药物靶点,利用已知肝癌数据库(Onco DB.HCC数据库和Liverome数据库)及GEO数据库筛选出乙型肝炎相关性肝癌的疾病靶点,用String构建蛋白质-蛋白质相互作用网络并筛选关键靶点,运用生物信息学方法对潜在靶点进行基因本体论(GO)分析和京都基因和基因组百科全书(KEGG)通路分析。利用GEPIA数据库对关键靶点进行生存预后分析。结果:获得柴胡疏肝散治疗乙型肝炎相关性肝癌的有效成分116个,潜在靶点17个,关键活性成分为槲皮素、山柰酚、β-谷甾醇、异鼠李素;关键靶点为雌激素受体1(Estrogen Receptor 1,ESR1)、细胞色素P4503A4酶(Cytochrome P450 Family 3 Subfamily A Member 4,CYP3A4)、醌氧化还原酶1[NAD(P)H Quinone Oxidoreductase 1,NQO1]。共涉及482个生物过程、5种细胞成分、57个分子功能,主要富集于12条信号通路。预测柴胡疏肝散主要通过干预类固醇激素的合成在乙型肝炎相关性肝癌的治疗中发挥重要的作用。结论:柴胡疏肝散通过多成分、多靶点、多途径治疗乙型肝炎相关性肝癌,且对抑郁也有干预作用,与中医疏肝解郁的治则相符。Objective:To explore the mechanism of Chaihu Shugan San(柴胡疏肝散)in the treatment of hepatitis B related hepatocellular carcinoma based on network pharmacology.Methods:The active components and drug targets of Chaihu Shugan San were screened by TCMSP platform.The disease targets of hepatitis B related hepatocellular carcinoma were screened by liver cancer database(OncoDB.HCC database and Liverome database)and GEO database.String was used to construct PPI network and screen key targets.The GO analysis and KEGG pathway analysis of potential targets were performed by bioinformatics methods.Survival analysis of key targets was performed using the GEPIA database.Results:Chaihu Shugan San has 116 active components and 17 potential targets in the treatment of hepatitis B related hepatocellular carcinoma.The key active components are quercetin,kaempferol,β-sitosterol and isorhamnetin,and the key targets are ESR1,CYP3A4 and NQO1.A total of 482 biological processes,5 cellular components and 57 molecular functions are involved,mainly enriched in 12 signal pathways.It is predicted that Chaihu Shugan San plays an important role in the treatment of hepatitis B related hepatocellular carcinoma mainly by interfering with the biosynthesis of steroid hormones.Conclusion:Chaihu Shugan San can treat hepatitis B related hepatocellular carcinoma through multi-component,multi-target and multi-pathway,and it also shows a certain effect on depression,which is consistent with the treatment principle of soothing the liver and relieving depression in traditional Chinese medicine.
