摘 要:研究旨在揭示蝾螈前梯度蛋白2(AGR2)基因的结构与功能,利用多种在线软件分析了5种蝾螈AGR2基因的生物信息学特征。结果显示:5种蝾螈AGR2基因的开放阅读框均为501 bp(编码166个氨基酸),编码产物分子量均为18 kD左右,理论等电点为8.87~9.20,不稳定指数均小于40,脂溶指数为93.37~102.77,平均亲水性指数为-0.368~-0.286。蝾螈AGR2具有一个信号肽,主要由α-螺旋和无规则卷曲构成,具有一个保守的硫氧还原蛋白类超家族结构域,其氨基酸序列与非洲爪蟾同源性最高。系统进化分析结果显示,蝾螈AGR2与非洲爪蟾AGR2亲缘关系最近。研究表明,蝾螈AGR2为可分泌蛋白,具有高度保守的硫氧还原蛋白类超家族结构域,与非洲爪蟾亲缘关系最近。The aim of the study was to reveal the structure and function of AGR2 gene in newts,and to analyze the bioinformatic characteristics of AGR2 gene in five species of newts using a variety of online software.The results showed that all the open reading boxes of the five newt AGR2 gene were 501 bp(encoding 166 amino acids),the molecular weight of the encoded products was about 18 kD,the theoretical isoelectric points were 8.87~9.20,the instability indexes were less than 40,the aliphatic indexes were 93.37~102.77,and the grand averages of hydropathicity were-0.368~-0.286.Newt AGR2 proteins had a signal peptide and mainly consisted ofα-helix and random coil,including a thiordoxin like superfamily domain.Their deduced amino acid sequences have the highest homology with that of Xenopus tropicalis.The phylogenetic analysis showed that newt AGR2 were closely related to that of Xenopus tropicalis.The study indicates that newt AGR2 proteins are secretory proteins with highly conserved thiordoxin like superfamily domains and had the closest relationship with Xenopus tropicalis.
