摘  要:细胞内pH对于维持细胞的正常运行以及生命系统的稳定具有重要作用。开发用于监测细胞内pH变化的新型荧光探针对于研究与其相关的疾病有着重要意义。本文以邻苯二胺和生物分子血红素为前驱体,通过一步水热法合成了铁掺杂荧光碳点(Fe-CDs)。Fe-CDs的制备方法简便,具有良好的水溶性和稳定的光学性能,高的荧光量子产率(26.80%),优异的抗盐及抗干扰能力。由于其表面丰富的氨基、羧基等官能团及铁元素的掺杂,Fe-CDs可以作为荧光探针用于酸性环境下的pH检测。研究表明,Fe-CDs的荧光强度在中性条件下最强,随着pH的减小逐渐降低。通过玻尔兹曼函数拟合曲线得到Fe-CDs的pKa为5.32,Fe-CDs的荧光强度在pH为4.4~6.2范围内具有良好的线性关系。因此,Fe-CDs可作为一种良好的pH荧光探针用于细胞内pH水平的监测,在生物传感和细胞成像方面具有应用潜力。Intracellular pH plays an important role in maintaining the normal state of cells,and its small fluctuations will have an important impact on biological systems.The development of new fluorescent probes for monitoring and measuring intracellular pH changes is of great significance for the study of related diseases.Using o-phenylenediamine and biological molecule hemin as precursors,we prepared the Fe-doped fluorescent carbon dots(Fe-CDs)through simply one-step hydrothermal method.Fe-CDs have good solubility,stable optical properties,high fluorescence quantum yield(26.80%),excellent salt resistance and anti-interference ability.Due to the abundant amino,carboxyl and other functional groups on its surface and the doping of iron,Fe-CDs can be used as fluorescent probes for pH detection in acidic environments.The results showed that the fluorescence intensity of the Fe-CDs reached the highest under neutral condition,and depressed gradually when the pH decrease.The fitted curve by boltzmann function showed that the pKa of Fe-CDs is 5.32.In the range of pH 4.4-6.2,the fluorescence intensity of Fe-CDs has a good linear correlation.Therefore,Fe-CDs can be used as a pH fluorescence probe for the monitoring of intracellular pH level and have potential application in biosensors and cell imaging.
