摘  要:紫花丹全草药用功能广泛,但其自然授粉率、种子萌发率和种苗存活率均非常低,自然种群分布越来越少。为保护紫花丹资源及满足未来的种苗需求,以紫花丹茎段为外植体,对其外植体消毒时间、芽增殖和壮苗生根培养基、移栽方式及基质进行研究。结果表明:利用0.1%氯化汞对紫花丹新生枝条消毒12 min效果较好;MS+6-BA 0.2 mg/L+IBA 0.1 mg/L可诱导茎段出芽,出芽后用MS+6-BA 2 mg/L+NAA 0.2 mg/L诱导芽增殖,增殖系数可达32.52;最佳生根培养基为1/2 MS+IBA 0.1 mg/L,15 d生根率可达61.57%;最佳移栽基质为泥炭土∶珍珠岩1∶1(V∶V),其生根率为76.67%,黑茎率最低(4.00%),新叶萌发率最高(78.00%);移栽方式以盖膜培养最佳,其植株新根及新叶萌发率最高,分别为76.67%和78%。本研究初步建立了紫花丹的离体快繁体系,为其工厂化育苗提供技术参考。The whole plant of Plumbago indica has many medicinal values.However,its fruiting rate of flowers that are left to be pollinated naturally,the germination rate of seed,and the survival rate of seedlings are very low,and its natural pop-ulation is getting smaller.To protect the resource of P.indica and meet the future demand of its seedling,stems segments of P.indica were used as explants to research the sterilization time for explants,the culture medium for the proliferation of shoots and induction of roots,transplanting pattern,and the transplant substrate in this research.The results show that the better method for the explants’disinfection is using 0.1%corrosive sublimate to soak for 12 minutes;buds can be inducted in the medium MS+6-BA 0.2 mg·L-1+IBA 0.1 mg·L-1,and shoots can be proliferated in the medium MS+6-BA 2 mg·L-1+NAA 0.2 mg·L-1(the multiplication coefficient can reach 32.52);the best culture medium for rooting is 1/2 MS+IBA 0.1 mg·L-1,in which the rooting rate reached 61.57%in 15 days;the best transplant medium is peat soil:perlite 1:1(V∶V)(the rooting rate is 76.67%,the rate of black stems is the lowest:4.00%and the germination rate of fresh leaves is the highest:78.00%);the best transplanting pattern is covering a film(the rooting rate and the germination rate of new leaves are the highest:76.67%and 78%respectively).This research has set up the preliminary rapid propagation system of P.indica and provides the theoretical foundation for its industrializing propagation.
