摘 要:目的探讨双夏秫米汤(半夏、夏枯草、薏苡仁1∶2∶1)对动物睡眠的改善作用。方法(1)采用回流提取法制双夏秫米汤浸膏,配置成高、中、低剂量(248.5、497、994 mg/kg)的混悬液,阳性药选用地西泮(0.9 mg/kg)首先分别以高、中、低剂量进行协同阈上剂量的戊巴比妥钠(35 mg/kg)翻正实验,观测不同剂量的双夏秫米汤对大鼠睡眠时长以及睡眠潜伏期的影响;(2)腹腔注射400 mg/(kg·d)对氯苯丙氨酸(P-chlorophenylalanine,PCPA)复制失眠模型大鼠。模型复制成功后分别予以高、中、低剂量的双夏秫米汤混悬液连续给药7天,治疗结束后采用协同阈上剂量的戊巴比妥钠翻正实验观测不同剂量的双夏秫米汤对失眠大鼠的睡眠时长和睡眠潜伏期的影响,再通过旷场实验分别记录各组大鼠在5分钟内的移动距离、穿格数、中央区停留时间、直立次数、修饰次数;(3)使用脑电图结合肌电图的方法分析大鼠灌胃高剂量双夏秫米汤后8小时内的睡眠结构,分别观测大鼠的睡眠潜伏期,苏醒时长、总睡眠时长、非快眼动睡眠时长、快眼动睡眠时长。结果(1)在协同阈上剂量的戊巴比妥钠翻正实验中双夏秫米汤高剂量组大鼠的睡眠时长显著延长,睡眠潜伏期显著缩短(P<0.05,P<0.05);(2)连续2天注射PCPA后造模大鼠表现出明显的攻击性,昼夜躁扰不宁,皮毛光泽较差并且食量下降,模型验证表明造模大鼠的睡眠潜伏期极为显著地延长(P<0.001),且睡眠时长非常显著地缩短(P<0.01),且经治疗后模型组大鼠的体重无明显增长(P<0.001),说明失眠模型复制成功。经治疗后各给药组大鼠的激惹性明显降低,双夏秫米汤中、高剂量组大鼠的睡眠潜伏期极为显著地缩短(P<0.001、P<0.001),高剂量组大鼠的睡眠时长非常显著地延长(P<0.01),且中、高剂量组大鼠的体重显著增长;在旷场实验中,双夏秫米汤高剂量组大鼠在旷场中的移动距离、穿格Objective To investigate the effect of the Shuangxia Shumi decoction for treating insomnia(Banxia,Xiakucao Yiyiren 1∶2∶1)on animal sleep.Methods(1)High,medium and low dose suspensions(248.5 mg/kg,497 mg/kg,994 mg/kg)of Shuangxia Shumi decoction were prepared by reflux extraction.First,the synergic upper threshold dose of pentobarbital orthosis experiment was conducted with high,medium and low doses,respectively.The sleep duration and sleep latency of rats were observed at different doses of Shuangxia Shumi decoction.(2)Rats with insomnia were induced by intraperitoneal injection of P-chlorophenylalanine(PCPA,400 mg/kg).After the model was successfully established,Shuangxia Shumi decoction suspension with high,medium and low doses was administered for 7 consecutive days,respectively.After the treatment,the effects of different doses of Shuangxia Shumi decoction on the sleep duration and sleep latency of rats with insomnia were observed by synergistic upper threshold pentobarbital transformation experiment.The open field experiment was used to record the movement distance,the number of penetration,the residence time in the central region,The times of standing up and the times of modification of rats in each group within 5 minutes.(3)Finally,electroencephalogram and electromyography were used to analyze the sleep structure of rats within 8h after gavage of high-dose Shuangxia Shumi decoction,and the sleep latency of rats was observed.The sleep latency,wake duration,total sleep duration,non-rapid eye movement sleep duration and rapid eye movement sleep duration of rats were observed.Results(1)Sleep duration was significantly extended and sleep latency was significantly shortened in high-dose Shuangxia shumi decoction group during the pentobarbital sodium experiment(P<0.05,P<0.05).(2)After two consecutive days of PCPA injection,the model rats showed obvious aggression,restless day and night,poor fur glossy and decreased food intake.Model verification showed that the sleep latency of the model rats was significantl
