摘  要:为了探究不同浓度维生素E在负压(-0.04 MPa)条件下对猪精液常温(17℃)保存效果的影响,试验将猪精液分为A~D组,分别在含0,0.1,0.5,1 mg/mL维生素E的Modena基础稀释液中保存,用全自动精子质量分析仪检测猪精子质量指标及畸形率,用相应试剂盒检测总抗氧化能力(T-AOC)及H_(2)O_(2)含量,用酸度计检测猪精液pH值。结果表明:在精液保存至第7,11天时,C组精子活率显著高于其他各组(P<0.05),且在保存至第1~5天时精子活率高于60%;在精液保存期间,C组精子活力高于其他组,且在第9天显著高于其他组;在精液保存期间,C组精子平均直线速度、平均曲线速度、平均路径速度及鞭打频率均高于其他组,畸形率低于其他组;在精液保存至第9天时,C组精子T-AOC显著高于其他组(P<0.05);在精液保存至第9,11天时,C组H_(2)O_(2)含量显著低于其他组(P<0.05);在精液保存期间,C组pH值的下降趋势相较其他组最缓慢,pH值最高,使猪精子处于较为适宜的弱酸环境中。说明将0.5 mg/mL维生素E添加到Modena基础稀释液中有助于猪精液在常温负压条件下保存。To investigate the effects of different concentrations of vitamin E on the preservation effects of porcine semen at room temperature(17℃)under negative pressure(-0.04 MPa),porcine semen was divided into groups A~D and was stored in Modena diluent containing 0,0.1,0.5 and 1 mg/mL vitamin E,respectively.The quality index and malformation rate of porcine sperm were detected by automatic sperm quality analyzer.The pH value of porcine semen was measured by acidity meter,and the total antioxidant capacity(T-AOC)and H_(2)O_(2) content were detected by corresponding kits.The results showed as follows:the sperm viability of group C was significantly higher than that of other groups(P<0.05)at the 7th and 11th day of semen preservation,and the sperm viability was higher than 60%on days 1-5.During semen preservation,the sperm motility in group C was higher than that in other groups,and significantly higher than that in other groups on day 9.During semen preservation,the mean linear velocity,mean curve velocity,mean path velocity and whip frequency in group C were higher than those in other groups,and the malformation rate was lower than that in other groups.On the 9th day of semen preservation,the T-AOC of sperm in the group C was significantly higher than that in other groups(P<0.05).The H_(2)O_(2) content in group C was significantly lower than that in other groups when semen was preserved until days 9 and 11(P<0.05).During the semen preservation,the decreasing trend of pH value in group C was the slowest compared with that in other groups,and the pH value was the highest,which made pig sperm in a more suitable weak acid environment.The results indicated that additng 0.5 mg/mL vitamin E to Modena basal diluent was helpful for the preservation of pig semen at room temperature under negative pressure.
