摘 要:甘薯茎尖脱毒技术是针对不同甘薯品种优化、种苗扩繁和防治多种病毒病的有效方法.试验以百薯2号为试验材料,分析不同外植体消毒方法和激素配比对于甘薯茎尖脱毒培养的影响.百薯2号茎尖脱毒培养的最佳外植体消毒方法为:体积分数为75%的乙醇溶液8 s+0.1%升汞6~7 min,污染率为12.5%;最佳茎尖诱导培养基为MS+1.0 mg/L 6-BA+0 mg/L NAA,平均诱导率为0.98±0.05;最佳再生苗增殖培养基为MS+1.0 mg/L 6-BA+0.1 mg/L NAA,平均成苗率为0.90±0.08.Virus-free tissue culture technology of sweet potato shoot tip is an effective method to optimize different sweet potato varieties,expand seedlings and prevent and control a variety of virus diseases.The effects of different explant disinfection methods and hormone concentration ratio on virus-free culture of sweet potato shoot tip were analyzed using Baishu 2 Hao.The best explant disinfection method for virus-free culture of Baishu 2 was 75%alcohol for 8 s+0.1%mercuric chloride for 6~7 min,and the pollution rate was 12.5%;The best shoot tip induction medium was MS+1.0 mg/L 6-BA+0 mg/L NAA,and the average induction rate was 0.98±0.05;The best regeneration medium was MS+1.0 mg/L 6-BA+0.1 mg/L NAA,and the average increment rate was 0.90±0.08.
