摘 要:目的:探究微小RNA 411(miR-411)靶向调节低氧诱导因子-1α(HIF-1α)对脊柱脊髓损伤(SCI)大鼠神经细胞凋亡的影响。方法:将44只大鼠随机分为假手术组、SCI组(模型组)、agomir-NC组、miR-411 agomir组;Allen′s法构建SCI模型;qRT-PCR检测脊髓组织中miR-411及HIF-1α表达。结果:相较于假手术组,SCI组脊髓组织中miR-411表达、Basso-Beattie-Bresnahan(BBB)评分降低,HIF-1αmRNA及蛋白表达、脊髓组织病理学程度、白细胞介素1β(IL-1β)及肿瘤坏死因子-α(TNF-α)水平、细胞凋亡指数、凋亡相关蛋白含半胱氨酸的天冬氨酸蛋白水解酶-3(Caspase-3)、凋亡诱导因子(AIF)表达增加(P<0.05);相较于SCI组和agomir-NC组,miR-411 agomir组miR-411表达、BBB评分增加,HIF-1αmRNA及蛋白表达、脊髓组织病理学程度、IL-1β及TNF-α水平、细胞凋亡指数、Caspase-3、AIF表达降低(P<0.05)。结论:miR-411过表达可能通过靶向抑制HIF-1α表达来抑制SCI后大鼠脊髓神经细胞凋亡。Objective:To explore the effect of microRNA 411(miR-411)on neuronal apoptosis in rats with spinal cord injury(SCI)by targeting hypoxia-inducible factor 1 alpha(HIF-1α).Methods:A total of 44 rats were randomly divided into sham operation group,SCI group(model group),agomir-NC group and miR-411 agomir group.Allen's method was used to construct SCI model;qRT-PCR was used to detect the expression of miR-411 and HIF-1αin spinal cord tissue.Results:Compared with that respectively in sham operation group,the miR-411 expression levels and the Basso-Beattie-Bresnahan(BBB)scores in the spinal cord tissue of the SCI group were significantly reduced,the HIF-1αmRNA and protein expression,spinal cord histopathology,interleukin 1β(IL-1β)and tumor necrosis factor-α(TNF-α)levels,apoptosis index,cysteinyl aspartate specific proteinase-3(Caspase-3),and apoptosis inducing factor(AIF)expression were increased(P<0.05);compared with that respectively in the SCI group/agomir-NC group,the miR-411 expression and BBB score in the miR-411 agomir group were increased,the HIF-1αmRNA and protein expression,spinal cord histopathologic scores,IL-1βand TNF-αlevels,apoptosis index,Caspase-3,and AIF expression were reduced(P<0.05).Conclusion:The overexpression of miR-411 may inhibit the apoptosis of spinal cord neurons in rats after SCI by targeting and inhibiting the expression of HIF-1α.
