摘  要:目的:观察按压耳穴刺激耳迷走神经对APP/PSl转基因阿尔茨海默病(AD)小鼠学习记忆能力的影响,从AD模型小鼠海马区谷氨酸(Glu)阳性细胞表达、突触数量与结构变化的角度探讨按压耳穴刺激耳迷走神经治疗AD的作用机制。方法:选取8个月龄AD双转基因APP/PS1小鼠12只,按随机数字表法将其分为模型组和治疗组,每组各6只;选取同背景同月龄C57BL/6J雄性小鼠6只作为对照组。治疗组按压小鼠耳穴“心”,压力控制在(350±50)g,每隔5s按压1次,总按压时间为15 min,1次/d,7 d为1个疗程,共干预4个疗程;对照组和模型组不予任何干预。干预结束后,采用Morris水迷宫检测各组小鼠学习记忆能力,免疫组化检测各组小鼠Glu阳性细胞的表达情况,采用透射电镜观察海马区突触数量与结构变化。结果:逃避潜伏期、目标象限时间、游泳总路程、穿台次数等指标,模型组与对照组、治疗组与模型组比较,差异均有统计学意义(P<0.05);小鼠海马CA1区Glu阳性细胞表达及其MOD,海马CA1区突触结构中PSD厚度、突触间隙、突触界面曲率等指标,模型组与对照组、治疗组与模型组比较,差异均有统计学意义(P<0.05)。结论:按压耳穴刺激耳迷走神经能够改善APP/PSl转基因AD模型小鼠的学习记忆能力,其作用机制可能与调控Glu水平、增强神经元突触可塑性有关。Objective:To investigate the effect of auricular point pressing for vagus nerve stimulation on the learning and memory abilities of APP/PSl transgenic mice with Alzheimer’s disease(AD),as well as its mecha-nism of action in the treatment of AD from the perspectives of the expression of Glu-positive cells and the number and structure of synapses in the hippocampus of AD mice.Methods:A total of 12 APP/PS1 transgenic mice,aged 8 months,were divided into model group and treatment group using a random number table,with 6 mice in each group;6 male C57BL/6J mice with the same age were selected as control group.The mice in the treatment group were given pressing at auricular point“Xin”with a pressure of 350±50 g,once every 5 seconds for a total pressing time of 15 minutes,once a day;each course of treatment was 7 days,and the mice were treated for 4 courses.The mice in the control group and the model group were not given any intervention.After intervention,the Morris water maze was used to observe learning and memory abilities;immunohistochemistry was used to measure the expression of Glu-positive cells;a transmission electron microscope was used to observe the change in the number and struc-ture of synapses in the hippocampus.Results:There were significant differences in escape latency,time spent in the target quadrant,total swimming distance,and the number of platform crossings between the model group and the control group and between the treatment group and the model group(P<0.05).There were also significant differ-ences in the expression and MOD of Glu-positive cells and related indices for synaptic structure(PSD thickness,synaptic cleft,curvature of synaptic interface)in the hippocampal CA1 region between the model group and the control group and between the treatment group and the model group(P<0.05).Conclusion:Auricular point press-ing for vagus nerve stimulation can improve the learning and memory abilities of APP/PSl transgenic mice with AD,possibly by regulating Glu level and enhancing neuronal synaptic
