摘 要:目的 探讨不同取血方式对流式细胞术测定小鼠外周血单核细胞亚群比例的影响。方法 18只6周龄雄性C57BL/6J小鼠,随机分为乙醚麻醉+下颌静脉丛取血组、下颌静脉丛取血组、乙醚麻醉+摘眼球取血组,每组6只。通过相应取血方式采取小鼠外周血,运用流式细胞术测定三组小鼠外周血单核细胞亚群比例。比较三组小鼠外周血Ly6Chi单核细胞亚群比例。结果 三组小鼠外周血Ly6Chi单核细胞亚群比例比较差异具有统计学意义(P<0.01)。乙醚麻醉+摘眼球取血组小鼠外周血Ly6Chi单核细胞亚群比例(60.1±1.4)%明显高于乙醚麻醉+下颌静脉丛取血组的(51.1±2.1)%,差异有统计学意义(P<0.01)。下颌静脉丛取血组小鼠外周血Ly6Chi单核细胞亚群比例(57.1±1.5)%高于乙醚+下颌静脉丛取血组的(51.1±2.1)%,但差异无统计学意义(P>0.05)。乙醚麻醉+摘眼球取血组与下颌静脉丛取血组小鼠外周血Ly6Chi单核细胞亚群比例比较差异无统计学意义(P>0.05)。结论 不同取血方式对小鼠外周血单核细胞亚群比例测定有差异,在涉及小鼠单核细胞亚群比例的研究中,应采取一致的取血方式。Objective To discuss the effects of different blood collection methods on the determination of the proportion of peripheral blood monocyte subsets in mice by flow cytometry.Methods A total of 18 male C57BL/6J mice aged 6 weeks old were randomly divided into ether anesthesia with mandibular venous plexus extraction group,mandibular venous plexus extraction group,ether anesthesia with eyeball extraction group,with 6 mice in each group.The peripheral blood of mice was collected by corresponding blood methods,and the proportion of peripheral blood monocyte subsets in the three groups was determined by flow cytometry.The proportion of Ly6Chi monocyte subsets in peripheral blood was compared among the three groups.Results The proportion of Ly6Chi monocyte subsets in peripheral blood of mice was significantly different among the three groups(P<0.01).The proportion of Ly6Chi monocyte subsets in peripheral blood of mice in the ether anesthesia+eyeball extraction group was(60.1±1.4)%,which was significantly higher than that of(51.1±2.1)%in the ether anesthesia with mandibular venous plexus extraction group,and the difference was statistically significant(P<0.01).The proportion of Ly6Chi monocyte subsets in peripheral blood of the mandibular venous plexus extraction group was(57.1±1.5)%,which was higher than that of(51.1±2.1)%of the ether anesthesiawith mandibular venous plexus extraction group,but the difference was not statistically significant(P>0.05).There was no statistically significant difference in the proportion of Ly6Chi monocyte subsets in peripheral blood between the ether anesthesia with eyeball extraction group and the mandibular venous plexus extraction group(P>0.05).Conclusion Different blood collection methods have differences in the determination of the proportion of peripheral blood monocyte subsets in mice,and the same blood collection method should be adopted in the experimental study of mouse monocyte subsets.
