摘 要:目的 探讨低密度脂蛋白受体(LDLR)、前蛋白转化酶枯草杆菌蛋白酶/kexin9(PCSK9)、肌球蛋白调控轻链结合蛋白(MYLIP)等10个基因的DNA甲基化与冠心病之间的相关性。方法 2012-2015年,在新疆医科大学第一附属医院招募35名冠心病患者作为病例组,35名正常者作为对照组。使用亚硫酸氢盐扩增子测序法(BSAS)对各个基因启动子区进行定量DNA甲基化分析,检测病例组与对照组基因CpG位点是否存在DNA甲基化的表达差异,同时检测具有差异的DNA甲基化单倍型。结果 本研究发现10个CpG位点DNA甲基化水平存在差异,26个单倍型存在差异,且差异均具有统计学意义(P<0.05)。结论 两组间脂代谢相关基因的DNA甲基化水平存在统计学差异,提示脂代谢相关基因的DNA甲基化在冠心病中起一定的调控作用。Objective To explore the correlation between DNA methylation of 10 genes,including LDLR,PCSK9,MYLIP and coronary heart disease.Methods From 2012 to 2015,35 patients with coronary heart disease were recruited as a case group and 35 normal patients as a control group at the hospital.Using bisulfite amplicon sequencing(BSAS)to quantitatively analyze DNA methylation in the promoter region of each gene,detecting whether there was a difference in the expression of DNA methylation at the CpG loci of the two groups of genes,and detecting the differenceDNA methylation haplotype.Results This study found that there were differences in DNA methylation levels of 10 CpG sites and 26 haplotypes,and the differences were statistically significant(P<0.05).Conclusion There was a statistical difference in the level of DNA methylation of lipid metabolism-related genes between the two groups,suggesting that the DNA methylation of lipid metabolism-related genes played a role in regulating coronary heart disease.
