重组人生长激素促进糖尿病模型大鼠创面愈合的机制研究

摘  要:目的探讨重组人生长激素(Recombinant human growth hormone,rhGH)促进2型糖尿病(T2DM)模型大鼠创面愈合的机制。方法35只Wistar大鼠中,选取28只建立T2DM模型,剩余7只正常大鼠作为对照组。35只大鼠共同建立背部创面,T2DM模型大鼠分为模型+盐水组(创周注射生理盐水)、GH2组(创周注射rhGH 0.2 IU/kg)、GH4组(创周注射rhGH 0.4 IU/kg)、GH6组(创周注射rhGH 0.6 IU/kg)。各组分别于干预第0、14 d取血,检测各组大鼠血清中表皮细胞生长因子(EGF)、酸性成纤维生长因子(aFGF)、血管内皮生长因子(VEGF)、胰岛素样生长因子(IGF)-1和血清白蛋白(ALB)水平。结果各组大鼠干预14 d后血清中aFGF(除模型+盐水组)、VEGF、EGF、IGF-1、ALB水平均较干预前升高,差异有统计学意义(P<0.05);干预后14 d,GH6组大鼠血清aFGF和ALB水平明显高于模型+盐水组、GH2和GH4组,差异有统计学意义(P<0.05),GH6组大鼠血清IGF-1水平明显高于模型+盐水组,差异有统计学意义(P<0.05)。结论rhGH可能是通过促进aFGF、IGF-1的表达和ALB的合成间接促进T2DM模型大鼠创面愈合。Objective To study the mechanism of recombinant human growth hormone(rhGH)on wound healing in type 2 diabetes(T2DM)rats.Methods 35 rats were randomly selected,among which 28 rats were established T2DM models,then the rest 7 normal rats were selected to establish the control group.35 rats were establish the back wound together.T2DM model rats were randomly divided into 4 groups:model+saline group(inject normal saline around periwound),GH2 group(inject rhGH 0.2 IU/kg around periwound),GH4 group(inject rhGH 0.4 IU/kg around periwound),GH6 group(inject rhGH 0.6 IU/kg around periwound).On 0 th and 14 th day of intervention,blood samples were taken from the each group to detect the levels of epidermal growth factor(EGF),acidic fibroblast growth factor(aFGF),vascular endothelial growth factor(VEGF),insulin-like growth factor(IGF-1),and serum albumin(ALB)in the serum of the rats in each group.Results After 14 days of intervention,the concentrations of aFGF(except model+saline group),VEGF,EGF,IGF-1 and ALB in the serum of the rats in each group were significantly increased than those before the intervention,the difference was statistically significant(P<0.05).After 14 days of the intervention,the serum levels of aFGF and ALB in GH6 group were significantly higher than those in model+saline group,GH2 and GH4 groups,the difference was statistically significant(P<0.05).The serum IGF-1 levels in GH6 group were significantly higher than those in model+saline group,the difference was statistically significant(P<0.05).Conclusion rhGH may indirectly promote wound healing in T2DM rats by promoting the expression of aFGF,IGF-1,and the synthesis of ALB.
