安胃疡胶囊促胃动力作用及防治慢性非萎缩性胃炎作用机制研究

摘　　要：目的 研究安胃疡胶囊对胃功能的影响及防治慢性非萎缩性胃炎的作用机制。方法 建立硫酸阿托品致胃动力不足小鼠模型,随机分为模型组(纯化水)、安胃疡低、中、高剂量组(安胃疡胶囊164、328、656 mg/kg体重)、胃乃安组(胃乃安740 mg/kg体重)、甲氧氯普胺组(甲氧氯普胺6 mg/kg体重),每天给药1次,每次0.2 mL/10 g体重,连续3 d,计算胃内残留率。建立大鼠慢性非萎缩性胃炎模型,随机分为模型组(纯化水)、安胃疡低、中、高剂量组(安胃疡胶囊82、164、328 mg/kg体重)、胃乃安组(胃乃安370 mg/kg体重)、甲氧氯普胺组(甲氧氯普胺3 mg/kg体重),每天给药1次,连续30 d。采用BL-420S生物机能实验系统测定胃收缩频率和收缩幅度;酸碱滴定法测定胃液酸度(包括游离酸及总酸度);pH计测定胃液pH值;胃蛋白酶(PP)试剂盒测定胃液PP活性;酶联免疫试剂盒(ELISA法)测定血清IL-8、GAS、MTL及胃组织IL-1β、PGE2水平;HE染色和AB-PAS染色观察胃组织、胃黏膜上皮细胞病理学变化。结果 安胃疡胶囊可明显降低胃动力不足小鼠的胃内残留率,与胃乃安组、甲氧氯普胺组比较,安胃疡高剂量组胃内残留率差异无统计学意义(P>0.05)。安胃疡胶囊可抑制慢性非萎缩性胃炎大鼠炎症、修复胃粘膜损伤、改善胃肠动力,与胃乃安组比较,安胃疡中、高剂量组胃组织病理学检查评分、胃黏膜上皮细胞粘蛋白指数、血清IL-8和MTL无明显变化。与甲氧氯普胺组比较,安胃疡高剂量组胃液量、pH值及安胃疡中、高剂量组血清GAS变化不大,差异均无统计学意义(P>0.05)。结论 安胃疡胶囊能够有效促进胃动力,防治慢性非萎缩性胃炎的发生。Objective To study the effect of Anweiyang capsuleon gastric function and its mechanism of prevention and treatment of chronic non-atrophic gastritis.Methods Mice with gastric motility insufficiency induced by atropine sulfate were randomly divided into model group(purified water),Anweiyang low,middle,high dose group(Anweiyang capsule 164,328,656 mg/kg body weight),Weinaian Group(Weinaian 740 mg/kg body weight)and metoclopramide group(metoclopramide 6 mg/kg body weight).The gastric residual rate was calculated after 3 days of administration with 0.2 mL/10 g once a day.Chronic non-atrophic gastritis rats were randomly divided into model group(purified water),Anweiyang low,middle,high dose group(Anweiyang capsule 82,164,328 mg/kg body weight),Weinaian group(Weinaian 370 mg/kg body weight)and metoclopramide group(metoclopramide 3 mg/kg body weight),administrated once a day at a dose of 1 mL/100 g body weight for 30 consecutive days.The gastric contraction frequency and amplitude was measured by BL-420S biological function experiment system;the acidity of gastric juice(including free acid and total acidity)measured by acid-base titration;pH value of gastric juice measured by pH meter;the activities of pepsin(PP)in gastric juice and the levels of IL-8,GAS,MTL in serum and IL-1β,PGE2 in gastric tissue determined by enzyme-linked immunosorbent assay(Elisa);HE staining and AB-PAS staining used to observe the changes of gastric cytopathology.Results Anweiyang capsule obviously reduced gastric residual rate in mice with gastric motility insufficiency,no significant difference showed in gastric residual rate between the high-dose group,Weinaian group and metoclopramide group(P>0.05).Anweiyang capsule could inhibit inflammation,repair gastric mucosa damage and improve gastrointestinal motility in chronic non-atrophic gastritis rats.Compared with Weinaian group,the middle,high-dose group of Anweiyang showed no obvious changes in gastric histopathology scores,mucin index of gastric mucosa epithelial cells and serum IL-8 and M
