摘  要:为了对陕西省和辽宁省部分地区羊源蜱类及其携带病原体的流行情况进行分析,本试验采集了288份羊源蜱样本,采用形态学观察鉴定蜱种;采用分子生物学方法扩增巴贝斯虫18S rRNA、伯氏疏螺旋体16S rRNA、斑点热群立克次体ompA和无形体16S rRNA的基因序列,并结合核苷酸序列分析,确定所收集蜱样本中各类病原体的流行情况。结果显示,经鉴定288份羊源蜱样本均为长角血蜱。陕西省144份长角血蜱中携带巴贝斯虫、斑点热群立克次体和无形体,感染率分别为0.69%、21.53%和77.78%,且巴贝斯虫和斑点热群立克次体均与无形体复合感染;其中长角血蜱中携带Babesia microti、Candidatus Rickettsia longicornis、Uncultured Rickettsia sp.QH-122、Rickettsia endosymbiont of Haemaphysalis longicornis、Uncultured Anaplasma sp.ZJ06/2009、Anaplasma marginale、Anaplasma capra共7种病原体。辽宁省144份长角血蜱中携带斑点热群立克次体和无形体,感染率分别为28.47%和93.06%,且斑点热群立克次体均与无形体复合感染;其中长角血蜱中携带Rickettsia raoultii、Rickettsia endosymbiont of Haemaphysalis longicornis、Anaplasma ovis、Anaplasma bovis共4种病原体。结果表明,陕西省和辽宁省部分地区羊源流行蜱种为长角血蜱,其携带多种病原体,且存在复合感染情况,本结果为防控蜱传疾病提供数据支持。In order to analyze the prevalence of ticks and tick-borne pathogens in sheep in some regions of Shaanxi and Liaoning provinces,a total of 288 tick samples from sheep were collected in this study.Morphological observation was used for tick species identification,and molecular biology methods were employed to amplify the gene sequences of Babesia 18S rRNA,Borrelia burgdorferi 16S rRNA,spotted fever group rickettsia ompA and Anaplasma 16S rRNA,and combined with nucleotide sequence analysis to determine the prevalence of various pathogens in the collected tick samples.The results showed that all 288 tick samples were identified as Haemaphysalis longicornis(H.longicornis).Babesia,spotted fever group rickettsia and Anaplasma were found in 144 H.longicornis from Shaanxi province,with the infection rate of 0.69%,21.53%and 77.78%,respectively;Babesia and spotted fever group rickettsia were found to be co-infected with Anaplasma;among the H.longicornis,a total of seven pathogens were detected,including Babesia microti,Candidatus Rickettsia longicornis,Uncultured Rickettsia sp.QH-122,Rickettsia endosymbiont of H.longicornis,Uncultured Anaplasma sp.ZJ06/2009,Anaplasma marginale and Anaplasma capra.Spotted fever group rickettsia and Anaplasma were found in 144 H.longicornis from Liaoning province,with the infection rates of 28.47%and 93.06%,respectively;and Babesia and spotted fever group rickettsia were found to be co-infected with Anaplasma;among the H.longicornis,a total of four pathogens were detected,including Rickettsia raoultii,Rickettsia endosymbiont of H.longicornis,Anaplasma ovis and Anaplasma bovis.The results indicate that the prevalent tick species in the selected regions of Shaanxi province and Liaoning province are H.longicornis,which carrying multiple pathogens and exhibiting co-infection.These findings provide data support for the prevention and control of tick-borne diseases.
