绵羊痘种毒保存期试验

摘　　要：鸡胚化绵羊痘羊体反应细胞毒(以下简称“绵羊痘种毒”)是制备绵羊痘活疫苗的基础种子,对于预防和控制绵羊痘疫情具有重要意义,定期对其检定十分必要。2000年版《中华人民共和国兽用生物制品规程》(以下简称“《规程》”)规定:绵羊痘种毒在-30℃以下,保存期为10年。为进一步探究绵羊痘种毒的保存期,本试验选取1981年1月、1983年1月、1990年4月和2006年2月冻干并于-70℃保存至今的4株绵羊痘种毒,进行病毒含量测定和最小发痘量测定。结果显示,4株绵羊痘种毒的最小发痘量均达到10^(-5)/0.5 mL,符合《规程》之规定;病毒含量分别为10^(3.3)、10^(3.8)、10^(4.8)和10^(5.5) TCID_(50)/0.1 mL,且与最小发痘量在一定范围内呈正相关。结果表明,冻干绵羊痘种毒在-70℃条件下保存39年仍符合生产活疫苗的免疫原性标准,因此可将绵羊痘种毒的保存期延长至-70℃保存30年,本试验为绵羊痘种毒的保藏检定和疫苗生产提供数据支撑。Sheeppox virus(SPPV)vaccine strain attenuated in chick embryo at different passage levels represents a valuable vaccine candidate,and infection of ovine testicular cells with attenuated sheeppox virus strain facilitates production of live vaccine strain.Given the significance of SPPV to prevent and control the outbreak of sheep pox,it is necessary to periodically test the efficacy of preserved virus seeds.The 2000 Version Veterinary Biological Products Regulations of the People's Republic of China defines the SPPV vaccine strain's preservation period as ten years at-30℃.To verify the preservation period of SPPV vaccine strain,four SPPV vaccine strains freeze-dried in January 1981,January 1983,April 1990 and February 2006,and preserved at-70℃,were selected for virus titer and minimum dilution inducing skin eruption determination.The results showed that the minimum dilution inducing skin eruption of all four SPPV vaccine strains reached 10-5/0.5 mL,which fully met the standards of the 2000 Version Veterinary Biological Products Regulations of the People's Republic of China.The viral titer of the above four SPPV vaccine strains were 103.3,103.8,104.8 and 105.5 TCID 50/0.1 mL,respectively,which were positively correlated with the minimum dilution inducing skin eruption within a certain range.The results indicate that freeze-dried SPPV vaccine strain stored at-70℃for 39 years still met the immunogenicity standards for live vaccine production.Therefore,the preservation period of SPPV vaccine strain can be extended to 30 years at-70℃.This study provides data support for the preservation,verification and vaccine production of SPPV vaccine strain.
