摘 要:为建立彩叶矾根‘栀子黄’的组培再生体系,了解其耐盐范围,以矾根‘栀子黄’的叶片为外植体,对其组培苗诱导培养基、耐盐范围和生根培养基等进行了筛选试验。结果表明:‘栀子黄’叶片最佳诱导培养基为MS+30 g·L^(-1)蔗糖+7 g·L^(-1)琼脂+0.2 mg·L^(-1)6-BA+0.01 mg·L^(-1)NAA,不定芽的诱导率达98.32%。耐盐性方面,0.2%轻度盐胁迫下,‘栀子黄’组培苗的存活率为92.45%,且该浓度对‘栀子黄’的长势具有一定的促进作用;在0.4%中度盐胁迫下,其存活率也可以达到75%,但是组培苗的再生率明显下降,玻璃化现象较重;在0.6%重度盐浓度下,组培苗成活率为35%,大部分组培苗褐化枯黄。‘栀子黄’未经盐胁迫处理组培苗的适宜生根培养基为MS+30 g·L^(-1)蔗糖+7 g·L^(-1)琼脂+0.05 mg·L^(-1)NAA,生根率为91.25%;耐盐组培苗的适宜生根培养基为MS+30 g·L^(-1)蔗糖+7 g·L^(-1)琼脂+0.1 mg·L^(-1)NAA,生根率为96.25%。试验结果可以为彩叶矾根种质资源的保存和工厂化育苗效率的提高提供参考,也可以为其在盐碱城市的绿化应用和耐盐机理的研究奠定基础。In order to establish tissue culture regeneration system for Heuchera micrantha‘Pistache'and understand its salt tolerance range,the experiment was conducted on the induction medium,salt tolerance range,and rooting medium of seedlings in vitro using the leaves as explants.The results showed that the best induction medium for‘Pistache'leaves was MS+30 g·L^(-1)sucrose+7 g·L^(-1)agar+0.2 mg·L^(-1)6-BA+0.01 mg·L^(-1)NAA,and the induction rate of adventitious buds had reached 98.32%.In terms of salt tolerance,the survival rate of seedlings in vitro was 92.45%at mild salt stress of 0.2%,and the growth had been promoted;the survival rate reached 75%at moderate salt stress of 0.4%,but the regeneration rate of seedlings in vitro was significantly decreased and the vitrification phenomenon was more severe;At severe salt concentration of 0.6%,the survival rate of seedlings in vitro was 35%,and most of seedlings in vitro turned brown and withered.The suitable rooting medium for seedlings of in vitro of‘Pistache'without salt stress was MS+30 g·L^(-1)sucrose+7 g·L^(-1)agar+0.05 mg·L^(-1)NAA,and the rooting rate was 91.25%;The suitable rooting medium for seedlings of in vitro of‘Pistache'with salt-tolerant was MS+30 g·L^(-1)sucrose+7 g·L^(-1)agar+0.1 mg·L^(-1)NAA,and the rooting rate was 96.25%.The experimental results will provide reference for the preservation of germplasm resources and the improvement of factory seedling efficiency of colored leaf alum root,and also lay a foundation for landscape applications in saline alkali cities and salt tolerance mechanism research.
