摘 要:为鉴定鸡白痢沙门菌生物被膜形成不依赖Rpo S的调控基因,以鸡白痢沙门菌S6702及其rpoS基因缺失株S6702ΔrpoS为母本,对S6702生物被膜状态和S6702ΔrpoS生物被膜状态(S7BF vs S7SBF)及S6702ΔrpoS浮游状态和S6702ΔrpoS生物被膜状态(S7SFY vs S7SBF)进行转录组测序分析,筛选差异表达基因,构建基因缺失株并验证其生物被膜形成能力。结果显示,S7BF vs S7SBF组共鉴定出83个差异表达基因,S7SFY vs S7SBF组共鉴定出1984个差异表达基因,筛选出15个可能与生物被膜形成相关的基因。以STM2361和STM1703为靶标构建基因缺失株,结晶紫染色定量结果显示,STM2361不影响鸡白痢沙门菌生物被膜的形成,而STM1703缺失能增强鸡白痢沙门菌的生物被膜形成能力。本研究鉴定出不依赖RpoS的鸡白痢沙门菌生物被膜形成调控基因,丰富了鸡白痢沙门菌生物被膜调控的信息。To identify the biofilm-forming related genes independent of Rpo S in Salmonella pullorum,Salmonella pullorum strain S6702 and its rpo S deletion strain S6702Δrpo S were selected as parent strains to perform transcriptome analyses of the paired group S6702 biofilm state vs S6702Δrpo S biofilm state(S7BF vs S7SBF) and the paired group S6702ΔrpoS planktonic state vs S6702ΔrpoS biofilm state(S7SFY vs S7SBF).Differentially expressed genes(DEGs) related to biofilm formation were selected to construct gene deletion mutants and the biofilm-forming ability of these mutants were determined.Results showed that 83 DEGs were discovered in the paired group S7BF vs S7SBF,1984 DEGs were discovered in the paired group S7SFY vs S7SBF,and 15 DEGs related to biofilm formation were screened out in these two groups.The STM2361 and STM1703 were selected to construct deletion mutants,crystal violet staining and quantification tests showed that the deletion of STM2361 had no effects on biofilm formation while deletion of STM1703 enhanced biofilm formation of Salmonella pullorum.Therefore,this study identifies several genes related to biofilm formation independent of Rpo S and enriches the information for regulation of biofilm formation in Salmonella pullorum.
