摘  要:由于具有高溶胀性,聚丙烯酰胺水凝胶有望用作胃滞留给药载体。然而水凝胶的力学性能在溶胀后会大幅度下降,使其不具备抵抗胃收缩载荷的能力,成为阻碍其临床应用的瓶颈问题。为实现聚丙烯酰胺水凝胶的稳定胃滞留,研究了交联剂浓度对水凝胶溶胀后力学性能的影响规律。结果表明,当交联剂浓度在0.05%~10%时,随着交联剂浓度的增加,水凝胶的平衡溶胀度下降,溶胀后的力学性能提高。当总聚合物浓度为20%,交联剂浓度为2%时,水凝胶的平衡溶胀度为7.5,溶胀后聚合物网络体积分数为9.0%,杨氏模量为86.5 kPa,具备用作胃滞留药物载体的潜力。Owing to its high swelling properties,polyacrylamide hydrogel has the potential to be used as drug carrier for gastric retention.However,the mechanical properties of hydrogel will be greatly reduced after swelling and the hydrogel cannot resist the gastric contraction,preventing its clinical applications.To achieve stable gastric retention of polyacrylamide hydrogel,the effect of concentration of cross-linker on the mechanical properties of the hydrogel after swelling was investigated.The results indicate that the equilibrium swelling degree of the hydrogel decreases and the mechanical properties after swelling are improved with the increase of cross-linker concentration in the range of 0.05%-10%.When the cross-linker concentration is 2%,the equilibrium swelling degree of the hydrogel is 7.5.The polymer network volume fraction after swelling is 9.0%,and Young’s modulus is 86.5 kPa.This hydrogel is suitable as the drug carrier for gastric retention.
