摘 要:为进一步解析鲜食葡萄种质的遗传关系及遗传多样性,为新品种选育提供理论依据。基于SCoT分子标记对泰安金牛山基地和郑州果树研究所国家葡萄种质资源圃收集的37份葡萄种质(优系)进行遗传关系及多样性调查,并利用NTSYS 2.10软件进行聚类和主坐标分析。结果表明,11对SCoT引物共扩增出115条清晰的谱带,平均每条引物扩增条带数为10.45条;115条谱带中多态性条带102条,平均多态性比例为87.90%;遗传相似系数分析显示其遗传范围在0.612~0.929;当遗传相似系数为0.67时,可将37份葡萄种质分为两大类,说明37份葡萄种质遗传背景存在差异,但多数葡萄种质与亲本或潜在亲本分布在同一类内。以上结果表明,SCoT分子标记可以分析葡萄种质的潜在亲本关系,该结果可用于葡萄育种的早期鉴定,为后期优良种质的选育提供理论依据。In order to further analyze the genetic relationship and diversity of table grape germplasm,and provide theoretical basis for new variety breeding.The genetic relationship and diversity of 37 grape germplasms from Taian Jinniushan Base and Zhengzhou National Grape Germplasm Nursery were analyzed using start codon targeted(SCoT)markers,cluster analysis and principal coordinate analysis were carried out by NTSYS2.10 software.The results showed that a total of 115 bands were amplified with 11 SCoT-PCR primers,with an average of 10.45 bands amplified by each primer,and the polymorphism ratio was 87.90%;the genetic similarity coefficient analysis of 37 grape germplasms showed that the average range was from 0.612 to 0.929.When the genetic similarity coefficient was 0.67,37 grape germplasms could be divided into two clusters,indicating that 37 grape germplasms have different backgrounds,but most grape accessions were distributed within the same cluster as the parent or potential parent.Therefore,all results show that SCoT markers can analyze the potential parental relationship of grape germplasm,which can be used for the early analysis of grape breeding.
