摘 要:基于米槠全基因组数据,利用生物信息学方法对米槠MADS-box基因家族进行鉴定,并对其系统进化关系、保守结构域、基因结构、基因复制和启动子顺式作用元件进行分析。此外,根据转录组数据构建米槠MADS-box基因在雄花序、雌花序、果实和果苞的不同发育阶段表达模式。本试验共鉴定出61个米槠MADS-box成员,所有基因均含有1个保守的MADS域,该域有保守的9段肽基序:KRK/R(X)4KK。系统进化和基因结构分析表明,有18个Type-Ⅰ型MADS-box基因,43个Type-Ⅱ型MADS-box基因,相同亚家族的成员有相似的基因结构和保守基序组成。61个成员不均等分布在12条染色体上,存在14对串联重复基因。表达量热图表明,Type-Ⅱ型A、B、C/D、E和AGL20类基因参与了雌花序的发育,B、C/D、E和AGL6类基因参与了雄花序的发育,E类基因可能参与雌花序休眠,而A类基因可能在雌花序解除休眠后发挥重要作用。A、C/D、E和AGL6类基因参与了果苞的发育,果苞可能由雌花的花器官发育而来。Type-Ⅱ型MADS-box基因家族成员在幼果期高表达,表明其参与果实早期发育。米槠FLC亚家族的CcMADS25可能在果实成熟过程中扮演重要角色。Genome-wide identification of MADS-box gene family was performed in Castanopsis carlesii for the analyses of their phylo-genetic relationship,conserved domain,gene structure,replication and cis-acting elements by bioinformatics methods.Additionally,the expression patterns of MADS-box gene family in different parts(male inflorescence,female inflorescence,fruit and bract)were further interpreted at different stages based on the transcriptome data.A total of 61 MADS-box genes were identified,and all of them had a conserved MADS domain containing a conserved motif(KRK/R(X)4KK).The phylogenetic tree demonstrated that 18 and 43 MADS-box genes were respectively classified into Type-Ⅰand Type-Ⅱ,and the genes in the same type shared similar structures and conserved motifs.The MADS-box genes were distributed on 12 chromosomes and comprised 14 pairs of tandem repeats.Transcriptom-ic analysis showed that the Type-Ⅱgenes of A,B,C/D,E and AGL20 participated in the development of female inflorescence,while B,C/D,E,and AGL6 genes were involved in the development of male inflorescence.Besides,E genes may play a role in the dormant process of female inflorescence,while A genes may be involved in its dormancy release.A,C/D,and AGL6 genes were in-volved in the development of the cupula.Type-ⅡMADS-box genes were highly expressed at the early fruit ripening stage,indicating its function at this stage.Furthermore,Cc MADS25 of the C.carlesii FLC subfamily may also play an important role in the process of fruit ripening.
