基于文献和系统药理学的黄连解毒汤治疗溃疡性结肠炎药效物质基础及其作用机制研究

摘　　要：目的:探讨黄连解毒汤(HLJDD)治疗溃疡性结肠炎(UC)的药效物质基础及其作用机制。方法:通过查阅文献,检索TCMSP、DrugBank、OMIM等数据库筛选HLJDD潜在活性成分及其作用靶点和UC疾病靶点,构建相关网络并借助拓扑学原理筛选HLJDD治疗UC的核心活性成分及核心作用靶点,并进行富集分析和分子对接验证。结果:获得HLJDD治疗UC核心活性成分6个(京尼平苷、小檗碱、巴马汀等),核心作用靶点360个。富集分析表明,HLJDD主要通过作用于NTRK1、CUL3、TP53等核心靶点,干预NF-κB、MAPK、Hippo等信号通路,进而调控细胞周期、DNA修复、细胞凋亡等生物过程来发挥治疗作用。结论:此研究从整体角度揭示了HLJDD治疗UC的潜在药效物质基础及其作用机制,可为HLJDD复方二次开发及抗溃疡新药研发奠定基础。Objective:To investigate the pharmacodynamic basis and mechanism of Huanglian Jiedu decoction(HLJDD)in the treatment of ulcerative colitis(UC).Methods:TCMSP,DrugBank,OMIM and other databases and literature review methods were used to screen the potential active components of HLJDD and their corresponding potential targets,as well as the disease targets of UC.Subsequently,the relevant network was constructed and the network topology analysis was used to screen the core active components and core targets of HLJDD in treating UC.Finally,enrichment analysis and molecular docking verification were performed.Results:Six core active components(geniposide,berberine,palmatine,etc.)and 360 core action targets of HLJDD in treating UC were obtained.Besides,enrichment analysis showed that HLJDD mainly interfered with NF-κB,MAPK,Hippo and other signaling pathways by acting on the core targets such as NTRK1,CUL3 and TP53,and then regulated cell cycle,DNA repair,apoptosis and other biological processes to play a therapeutic role.Conclusion:The study revealed the potential pharmacodynamic material basis and mechanism of HLJDD in treating UC from the overall perspective,which can provide basis for clinical treatment of UC by HLJDD,and also lay a foundation for the secondary development of HLJDD prescription and development of new anti-ulcer drugs.
