摘 要:目的观察柴芍承气汤对脓毒症大鼠胰腺损伤的影响,探讨其可能的作用机制。方法36只Wistar大鼠随机分为对照组、脓毒症组和柴芍承气汤组,脓毒症组和柴芍承气汤组采用盲肠结扎穿孔法建立脓毒症模型,柴芍承气汤组于造模前1 h及造模后6 h各灌胃1次。生化法检测大鼠胰腺组织髓过氧化物酶(MPO)、脂质过氧化物(LPO)、谷胱甘肽(GSH)水平,免疫组化和Western blot检测胰腺组织半胱氨酸白三烯受体1(CysLTR1)、核因子(NF)-κB蛋白表达,TUNEL染色检测胰腺组织细胞凋亡情况。结果与对照组比较,脓毒症组大鼠胰腺组织MPO、LPO水平显著升高,GSH水平显著降低(P<0.001),CysLTR1、NF-κB蛋白表达显著升高(P<0.01),细胞凋亡率显著升高(P<0.001);与脓毒症组比较,柴芍承气汤组大鼠胰腺组织MPO、LPO水平显著降低,GSH水平显著升高(P<0.05,P<0.01),CysLTR1、NF-κB蛋白表达显著降低(P<0.05),细胞凋亡率显著降低(P<0.01)。结论柴芍承气汤对脓毒症大鼠胰腺损伤具有一定保护作用,其机制与抑制细胞凋亡,下调CysLTR1介导的NF-κB信号通路有关。Objective To investigate the effects of Chaishao Chengqi Decoction on pancreatic injury in septic rats;To explore its possible mechanism.Methods Totally 36 Wistar rats were randomly divided into control group,sepsis group and Chaishao Chengqi Decoction group,the sepsis group and Chaishao Chengqi Decoction group used cecal ligation and puncture to establish sepsis model,the Chaishao Chengqi Decoction group was administered by gavage 1 hour before modeling and 6 hours after modeling.The levels of myeloperoxidase(MPO),lipid peroxide(LPO)and glutathione(GSH)in pancreatic tissue were detected by biochemical method,the protein expressions of cysteine leukotriene receptor 1(CysLTR1)and nuclear factor(NF)-κB in pancreatic tissue were detected by immunohistochemistry and Western blot,TUNEL staining was used to detect apoptosis in pancreatic tissue.Results Compared with the control group,the levels of MPO and LPO in pancreatic tissue of the sepsis group significantly increased,the level of GSH significantly decreased(P<0.001),the expressions of CysLTR1 and NF-κB protein significantly increased(P<0.01),and the apoptosis rate significantly increased(P<0.001).Compared with the sepsis group,the levels of MPO and LPO in pancreatic tissue of rats in Chaishao Chengqi Decoction group significantly decreased,the level of GSH significantly increased(P<0.05,P<0.01),the expressions of CysLTR1 and NF-κB protein decreased significantly(P<0.05),and the apoptosis rate decreased significantly(P<0.01).Conclusion Chaishao Chengqi Decoction has a certain protective effect on pancreatic injury in septic rats,and its mechanism is related to inhibiting cell apoptosis and down-regulating the NF-κB signaling pathway mediated by CysLTR1.
