摘  要:丝腺是家蚕合成和分泌丝蛋白的唯一器官,温度在丝腺合成丝蛋白的过程中具有重要调控作用,对蚕丝产量和品质都有重要影响。了解丝腺对饲养温度的响应机制对有效调控熟蚕吐丝有重要借鉴作用,以实用蚕品种贵蚕7号为研究对象,在丝腺发育和丝蛋白合成关键时期(5龄期)分别采用20℃、25℃和30℃不同温度条件进行饲养,并取游走期丝腺进行转录组比较分析。结果发现高温组(30℃)和低温组(20℃)与正常适宜饲养温度组(25℃)相比,共鉴定到109053个差异表达基因。其中30℃饲养条件下存在67个显著上调表达基因,196个显著下调表达基因;20℃饲养条件下存在101个显著上调表达基因,124个显著下调表达基因。高温组和低温组之间分别存在4个和7个特异差异表达基因。进一步对这些差异基因进行GO和KEGG功能富集分析,发现高温组差异表达基因主要集中于氨基酸代谢、保幼激素代谢和能量代谢等与丝蛋白合成紧密相关的信号通路,低温组差异表达基因主要富集于苯丙素的生物合成、谷胱甘肽代谢等与物质代谢合成相关的信号通路,共同富集的通路主要集中于物质运输和能量代谢。这说明高温和低温胁迫影响丝腺发育或丝蛋白合成分泌的分子途径既存在显著差异又有部分相似之处,高温可能同时影响到丝腺的发育和丝蛋白的合成来影响蚕丝产量,而低温可能主要是通过能量代谢间接影响丝蛋白的合成。上述结果为理解家蚕丝腺对饲养温度的响应机制提供了参考依据。Silk gland is the unique tissue for silkworm to synthesize and secrete silk proteins.Temperature plays an important regulatory role in the process of silk protein synthesis in silk gland,and has an important impact on silk yield and quality.Understanding the response mechanism of silk gland to feeding temperature could provide good references to control the silk spinning of mature silkworms.The commercial silkworm variety Guican 7 was taken as the research object,and at the criticel stage of silk gland development and silk protein synthesis(the fith instar stage),different temperature conditions were used for feeding at 20℃,25℃ and 30℃ respectively.Then the transcriptome of silk gland at the wandering stage were compared and analyzed.The results showed that compared with the normal feeding temperature group(25℃),109053 differentially expressed genes were identified in the high temperature group(30℃)and the low temperature group(20℃).Among them,there were 67 significantly up-regulated genes and 196 significantly down-regulated genes under the feeding conditions of 30℃;There were 101 significantly upregulated genes and 124 significantly down-regulated genes under the feeding conditions of 20℃.There were 4 and 7 specific differentially expressed genes in the high temperature group(30℃)and the low temperature group(20℃)respectively.Further analysis of GO and KEGG function enricnment of these differential genes showed that in the high-temperature group they were mainly enriched in the signaling pathways closely related to silk protein synthesis such as amino acid metabolism,juvenile hormone metabolism and energy metabolism,while in the low-temperature group they were mainly enriched in the signaling pathways related to substance metabolism and synthesis,such as biosynthesis of pheny-Ipropyl and glutathione metabolism.And the co-enriched pathways were mainly enriched in material transport and energy metabolism.All these indicated that the molecular pathways of high temperature and low temperature st
