摘 要:Toll和IMD通路是昆虫先天免疫的重要信号通路,抗菌肽的产生可以由这些通路激活。通过感染4种不同种类的病原体(球孢白僵菌、大肠埃希菌、金黄色葡萄球菌和枯草芽孢杆菌),对家蚕Toll通路的BmTube、BmMyD88、BmTollip、BmECSIT、BmTRAF2和BmCactus基因,IMD通路的BmDredd、BmFadd、BmTAB2、BmTAK1、BmRelish1和BmRelish2基因以及4类不同抗菌肽(antimicrobial peptide,AMP)基因BmCecropinA1、BmAttacin1、BmDefensinA和BmGloverin1的表达进行分析,探讨家蚕在感染不同病原后Toll和IMD通路级联反应的免疫响应。结果表明,家蚕Toll和IMD信号通路级联反应基因表达调控与不同种类病原体感染相关;不同类型的病原选择性地激活Toll和IMD通路,进而诱导特定AMP基因的表达,AMP基因表达对细菌的免疫响应快,对真菌的免疫响应慢,其中BmCecropinA1表达强度显著高于其他类型AMP基因。Toll and IMD pathways are important signaling pathways of insect innate immunity,and the production of antimicrobial peptides(AMPs)can be activated by these pathways.After Bombyx mori was infected by four pathogens(Beauveria bassiana,Escherichia coli,Staphylococcus aureus and Bacillus subtilis),the expression analysis of BmTube,Bm-MyD88,BmTollip,BmECSIT,BmTRAF2 and BmCactus genes in Toll pathway,BmDredd,BmFadd,BmTAB2,BmTAK1,BmRelish1 and BmRelish2 genes in IMD pathway,and four AMP genes BmCecropinA1,BmAttacin1,BmDefensinA and BmGloverin1,was carried out to explore the immune response of the cascade reactions of Toll and IMD pathways in Bombyx mori after infection.The results showed that the regulation of gene expression in the cascade reactions of Toll and IMD pathways was related to the infection of different types of pathogens,which selectively activated the Toll or IMD pathway,and then induced the expression of specific AMP genes.The expression of AMP genes had a fast immune response to bacteria but a slow immune response to fungi.The expression intensity of BmCecropinA1 gene was much higher than that of other types of AMP genes.
