摘  要:目的:利用近红外光谱技术建立一种快速、准确且低损的鹿茸鉴别方法。方法:采集40份药典收载鹿茸(梅花鹿茸、马鹿茸)及32份未被药典收载鹿茸(驯鹿茸、花马鹿茸等)共72份鹿茸样品的近红外光谱数据,选取信息量丰富的10000~4000 cm^(-1)区段作为分析对象,原始数据经标准正态变量变换(SNV)、多元散射校正(MSC)、一阶导数(1stDer)、二阶导数(2ndDer)和Savitzky-Golay平滑(SG)等光谱预处理,结合化学计量学实现了不同鹿茸饮片的鉴别。结果:1stDer结合OPLS-DA可以鉴别药典收载鹿茸与药典未收载鹿茸;MSC结合OPLS-DA可以鉴别梅花鹿茸和马鹿茸;MSC+2ndDer结合OPLS-DA可以鉴别梅花鹿茸的不同等级,而马鹿茸用Raw即可鉴别不同的等级;梅花鹿茸的两个产地通过MSC结合OPLS-DA可以鉴别。结论:近红外光谱技术结合化学计量学不仅可用于快速鉴别鹿茸饮片的真伪,还可用于不同规格和等级的鹿茸品质评价,为鹿茸的质量控制提供了具开发潜力的快检新方法。Objective:To establish a rapid,accurate and low damage identification method for Cervi Cornu Pantotrichum by near infrared spectroscopy.Methods:The near infrared spectral data of 72 Cervi Cornu Pantotrichum samples were collected from 40 samples collected in pharmacopoeia(cornu cervi of Cervus nippon,cornu cervi of Cervus elaphus)and 32 samples not collected in pharmacopoeia(cornu cervi of Rangifer tarandus,crossed cornu cervi of Cervus Nippon and Cervus elaphus).The 10 000~4 000 cm^(-1) segment with abundant information was selected as the analysis object.The original data were preprocessed by the standard normal variable transformation(SNV),multiple scattering corrections(MSC),first derivative(1stDer),second derivative(2ndDer) and Savitzky-Golay smoothing(SG) spectra,and combined with stoichiometry to achieve the identification of different pieces.Results:1stDer combined with OPLS-DA could distinguish Cervi Cornu Pantotrichum collected in pharmacopoeia and not collected in pharmacopoeia.MSC combined with OPLS-DA could identify cornu cervi of Cervus nippon and Cervus elaphus.MSC+2ndDer combined with OPLS-DA could identify different grades of cornu cervi of Cervus nippon,while Raw could identify different grades of cornu cervi of Cervus elaphus.The two origins of cornu cervi of Cervus nippon were identified by MSC combined with OPLS-DA.Conclusion:Near infrared spectroscopy combined with chemometrics can be used for rapid identification and quality evaluation of Cervi Cornu Pantotrichum pieces,it provides a reference for the study of quality standard control of Cervi Cornu Pantotrichum.
