摘  要:目的:采用HPLC和UPLC-Q-TOF-MS/MS方法分析白及正丁醇提取物,与止血作用相关联,明确白及止血物质基础。方法:建立16批白及正丁醇提取物HPLC指纹图谱,以毛细管法和断尾法考察小鼠凝血和出血时间,评价止血作用;采用灰色关联度分析法、双变量相关分析法及偏最小二乘回归分析法研究谱效关系。结果:白及正丁醇提取物有15个共有峰,其中1、8、9、11、13~15号色谱峰与止血作用的关联度>0.6且Pearson相关系数为正,利用质谱数据结合对照品确定1号为天麻素、8号为gymnosideⅠ、9号为militarine,偏最小二乘回归分析确定1、4、8~10号色谱峰与止血时间呈负相关,且VIP值>1。结论:白及止血活性成分为gymnosideⅠ、militarine,该研究可为提升白及质量标准和进一步药效研究提供参考。Objective:To analyze the n-butanol extract of Bletillae Rhizoma by HPLC and UPLC-Q-TOF-MS/MS methods,to associated with hemostatic effect and clarify the hemostatic material basis of Bletillae Rhizoma.Methods:HPLC fingerprint of 16 batches of Bletillae Rhizoma n-butanol extracts was established,and the blood coagulation and bleeding times of mice were evaluated by capillary method and tail-breaking method.Grey correlation analysis,bivariate correlation analysis and partial least squares regression analysis were used to study the spectrum effect relationship.Results:There were 15 common peaks in the n-butanol extract of Bletillae Rhizoma,among which the correlation degree of peaks 1,8,9,11,13 to 15 with haemostatic action was greater than 0.6 and the Pearson correlation coefficient was positive.Using mass spectrometry data combined with reference substance,peak 1 was gastrodin,peak 8 was gymnoside Ⅰ and peak 9 was militarine.Partial least squares regression further analysis showed that peaks 1,4,8 to 10 were negatively correlated with hemostasis action and VIP value was greater than 1.Conclusion:The hemostatic active components of Bletillae Rhizoma are the chemical constituents represented by gymnoside Ⅰ and militarine.This study provides a scientific basis for improving the quality standard and further pharmacodynamic study of Bletillae Rhizoma.
