摘 要:目的考察迷迭香酸在大鼠离体小肠中的吸收机制。方法Ussing Chamber分析0.5、1、2、4 mg/mL迷迭香酸在肠黏膜中的透过情况,探讨P-糖蛋白抑制剂维拉帕米对该成分肠黏膜透过的影响,以及不同辅料(泊洛沙姆188、聚氧乙烯35蓖麻油、吐温80)对其小肠吸收的促进作用。结果迷迭香酸在大鼠十二指肠、空肠、回肠中的吸收速率与其质量浓度呈线性关系(r>0.90),加入维拉帕米后吸收提高(P<0.05)。3 mg/mL泊洛沙姆188、3 mg/mL聚氧乙烯35蓖麻油增加迷迭香酸在空肠、回肠中的吸收(P<0.05)。结论迷迭香酸在大鼠离体小肠中的吸收机制为被动扩散,其肠黏膜吸收过程受P-糖蛋白转运体影响,泊洛沙姆188、聚氧乙烯35蓖麻油可促进其吸收。AIM To investigate the absorption mechanism of rosmarinic acid in rat small intestine in vitro.METHODS Ussing Chamber was employed to analyze the intestinal mucosal permeability of 0.5,1,2,4 mg/mL rosmarinic acid,after which the effect of P-glycoprotein inhibitor verapamil on intestinal permeability of this constituent and promoting effect of different excipients(poloxamer 188,polyoxyethylene 35 castor oil,Tween 80)on its small intestinal absorption were explored.RESULTS The absorption rates of rosmarinic acid in rat duodenum,jejunum and ileum were linearly related to its concentration(r>0.90),whose absorption was enhanced after the addition of verapamil(P<0.05),3 mg/mL Poloxamer 188 and 3 mg/mL polyethylene glycol 35 castor oil increased the absorption of rosmarinic acid in jejunum and ileum(P<0.05).CONCLUSION The absorption mechanism of rosmarinic acid in rat small intestine in vitro is passive diffusion,whose intestinal mucosal absorption process is affected by P-glycoprotein transporter,poloxamer 188 and polyoxyethylene 35 castor oil can promote its absorption.
