摘　　要：目的观察钩藤降压解郁方对高血压并发抑郁大鼠血压及抑郁样行为的改善作用。方法40只自发性高血压大鼠(SHR),随机均分为模型组、阳性对照组和钩藤降压解郁方低、中、高剂量(6.345、12.69、25.38 g/kg)组,另取8只SD大鼠为正常组。以慢性不可预知性应激(CUMS)联合孤养大鼠的方法复制高血压并发抑郁模型。造模同时灌胃给药,每天1次,连续42 d。采用无创血压仪检测各组大鼠尾动脉血压,旷场实验、糖水消耗实验、强迫游泳实验检测各组大鼠总活动次数、糖水偏好率、强迫游泳不动时间,HE染色和尼氏染色观察海马区神经元的活性、排列情况以及神经元尼氏小体的数量,ELISA法检测大鼠海马组织单胺递质5-HT、NE、DA与炎性因子COX-2、MCP-1、TNF-α水平。结果造模42 d后,与模型组比较,阳性对照组和钩藤降压解郁方低、中剂量组尾动脉收缩压降低(P<0.01),阳性对照组和钩藤降压解郁方中、高剂量组旷场实验总活动次数、糖水消耗率升高(P<0.01),强迫游泳实验不动时间缩短(P<0.01),海马神经元细胞状态好转,排列整齐,少见凋亡细胞,尼氏小体丰富,海马组织中单胺递质5-HT、NE、DA水平升高(P<0.01),炎性因子COX-2、MCP-1、TNF-α分泌减少(P<0.05,P<0.01)。结论钩藤降压解郁方能通过减少海马组织炎性因子分泌,稳定单胺类神经递质表达,缓解海马神经元损伤,实现对高血压并发抑郁症大鼠血压及抑郁样行为的改善作用。AIM To observe the effects of Gouteng Jiangya Jieyu Recipe on blood pressure and depression-like behavior in hypertensive rats complicated with depression.METHODS Forty spontaneously hypertensive rats(SHR)were randomly divided into the model group,the positive control group,and the Gouteng Jiangya Jieyu Recipe groups for low,medium and high dose use(6.345,12.69,25.38 g/kg)repspectively,in contrast to other eight SD rats of the normal group.The rat models of hypertension complicated with depression were induced by chronic unpredictable stress(CUMS)combined with social isolation and treated orally with the corresponding agent once daily for 42 days.The rats had their blood pressure of the tail artery measured by non-invasive blood pressure meter;their total locomotor activity,sugar water preference rate and immobility/swim time detected by open field test,sucrose preference test and forced swimming test;their activity and arrangement of neurons in hippocampus and number of Nissl bodies observed by HE staining and Nissl staining;and their hippocampal monoamine transmitters 5-HT,NE,DA and inflammatory factors COX-2,MCP-1 and TNF-αlevels determined by ELISA.RESULTS After 42 days of modeling,compared with the model group,the positive control group and the low-dose and middle-dose groups of Gouteng Jiangya Jieyu Recipe displayed decreased systolic blood pressure in tail artery(P<0.01);the positive control group,the middle-dose and high-dose groups of Gouteng Jiangya Jieyu Recipe shared increased total locomotor activity and sugar water consumption rate(P<0.01),shortened immobility time in forced swimming experiment(P<0.01),abundant Nissl bodies,and increased levels of hippocampal monoamine transmitters 5-HT,NE and DA(P<0.01),and decreased secretion of inflammatory factors COX-2,MCP-1 and TNF-α(P<0.05,P<0.01).CONCLUSION Gouteng Jiangya Jieyu Recipe can improve the blood pressure and depression-like behavior in hypertensive rats complicated with depression by reducing the secretion of inflammatory factors in hippocampus
