摘 要:目的:采用经过改进与优化的高效液相色谱(HPLC)法对人体血清中的甲氨喋呤(MTX)血药浓度进行监测,评定其效能与价值。方法:采集血清样品100μL,用高氯酸溶液(15%)进行处理,以HPLC法实施测定。流动相:甲醇-10mmol/L磷酸二氢钾(20∶80,采用氢氧化钠调至pH6.5),色谱柱:大连依利特ODS C_(18),流速:1.0mL/min,紫外检测波长:303nm,柱温:35℃。结果:MTX的血药浓度位于0.1~64μmol/L这一范围内,会有较好的线性关系(r=0.9995)。MTX低浓度的日内、日间精密度(RSD)分别为2.69%、4.35%,而提取回收率是70.8%;中浓度分别是1.07%、2.70%与78.2%,高浓度则为0.93%、1.94%与75.3%;最低检测限是0.03μmol/L。结论:经过改建与优化后的HPLC法不仅准确、可靠,而且还有着较好的重现性,较高的灵敏度,在监测大剂量MTX的血药浓度当中,有着较好的应用价值。To use an improved and optimized high-performance liquid chromatography(HPLC)method to monitor the blood concentration of methotrexate(MTX)in human serum,and evaluate its efficacy and value.Method:Collect 100 serum samplesμL.Treat with a perchloric acid solution(15%)and perform the determination using HPLC.Mobile phase:Methanol-10mmol/L potassium dihydrogen phosphate(20∶80,adjusted to pH 6.5 with sodium hydroxide),chromatographic column:Dalian Yilite ODS C_(18),flow rate:1.0mL/min,UV detection wavelength:303nm,column temperature:35℃.Result:The blood drug concentration of MTX is between 0.1 and 64μWithin the range of mol/L,there will be a good linear relationship(r=0.9995).The intra day and inter day precision(RSD)of low concentration MTX were 2.69%and 4.35%,respectively,while the extraction recovery rate was 70.8%;The medium concentration is 1.07%,2.70%,and 78.2%,respectively,while the high concentration is 0.93%,1.94%,and 75.3%;The minimum detection limit is 0.03μmol/L。Conclusion:The modified and optimized HPLC method is not only accurate and reliable,but also has good reproducibility and sensitivity.It has good applicability in monitoring the blood concentration of high-dose MTX.
