摘  要:为筛选出与鸡抗病力相关的遗传标记,以便为家禽的抗病育种提供分子水平的理论基础。该文选取在相同饲养条件下、同为100日龄(免疫程序一致)的5个广西不同地方品种鸡(广西麻鸡、广西乌鸡、瑶鸡、龙胜凤鸡、七百弄鸡)各30只,经羽下静脉采血,应用Elisa方法检测鸡血清中干扰素α、β、γ含量,用荧光定量PCR方法检测不同品种鸡在基因水平上干扰素α、β、γ及干扰素上游受体蛋白黑色素瘤分化相关抗原5(MDA5)、下游抗病毒蛋白2’,5’寡腺苷酸合成酶(OAS)、Mx蛋白的表达水平差异。结果表明:七百弄鸡血清干扰素α、β、γ水平均显著高于其余品种,尤其是血清中干扰素γ含量为1823.33 pg/mL,约为乌鸡干扰素γ含量的5倍;荧光定量PCR结果显示不同品种鸡干扰素α、β、γ基因相对表达量差异显著,七百弄鸡干扰素基因相对表达量最高,MDA5、OAS、Mx蛋白呈现出一致的表达趋势。说明在相同生长环境条件下,七百弄鸡干扰素诱生水平不同程度地优于其他4个品种,七百弄鸡的抗病力存在某些优势。In order to screen out the genetic markers related to chicken disease resistance and provide the theoretical basis of molecular level for disease resistance breeding in poultry.In this paper,30 chickens from 5 different local chicken breeds(Guangxi ma chicken,Guangxi black chicken,Yao Chicken,longsheng chicken and Qibailong Chicken)of 100 days old were selected and fed under the same conditions,eLISA was used to detect the content of interferon-α,interferon-β,interferon-γin chicken serum,the expression levels of interferonα,β,γ,melanoma differentiation associated antigen 5(MDA5),downstream antiviral protein 2,5;oligoadenylate synthetase(OAS)and MX protein in different chicken breeds were detected by quantitative real-time PCR.The results showed that the levels of interferon-α,interferon-β,and interferon-γin serum of 700-nong chickens were significantly higher than those of other breeds,especially the level of interferon-γin serum was 1823.33 pg/mL,which was about five times of that of interferon-γin silky fowl.The results of quantitative real-time PCR showed that the relative expression levels of interferon-α,interferon-β,and interferon-γgenes in different breeds were significantly different.The relative expression levels of interferon-α,interferon-β,and interferon-γgenes in 700-nong chickens were the highest,and the expression trends of MDA5,OAS,and MX proteins were consistent.The results indicated that under the same growth environment,the level of interferon induction of 700-nong chicken was better than that of other four varieties in different degrees,and the disease resistance of 700-nong chicken had some advantages.
