摘  要:目的探讨β2肾上腺素受体(ADRB2)基因rs1042713、白细胞介素4受体(IL-4R)基因rs1801275位点的多态性与儿童哮喘的相关性。方法采用临床病例对照法,选取黑衣壮中年龄<16岁的儿童200例。其中哮喘组100例,健康组100例。采用聚合酶连接酶链式反应(PCR-LCR chain reaction)检测ADRB2基因rs1042713、IL-4R基因rs1801275位点多态性,并分析其检测结果。结果(1)ADRB2基因rs1042713、IL-4R基因rs1801275位点均检测出AA、AG、GG三种基因型;(2)ADRB2基因rs1042713、IL-4R基因rs1801275位点基因型在哮喘组与健康组中差异有统计学意义(P<0.05);(3)ADRB2基因rs1042713位点基因型AA、AG+AA和等位基因A可能增加哮喘发病风险,GG+AG可降低哮喘发病风险。IL-4R基因rs1801275位点AG、AG+GG和等位基因G可降低哮喘发病风险,相较杂合子AG,纯合子AA+GG能增加哮喘发病风险。结论(1)广西黑衣壮儿童中存在ADRB2基因rs1042713、IL-4R基因rs1801275位点多态性;(2)ADRB2基因rs1042713、IL-4R基因rs1801275位点可能是广西黑衣壮儿童哮喘发病的相关因子。Objective To investigate correlation between the polymorphisms of the rs1042713 of Beta-2 adrenergic receptor(ADRB2)gene and the rs1801275 of interleukin-4 receptor(IL-4R)gene and asthma of Heiyi Zhuang children in Guangxi.Methods Clinical case-control method was used to selected 200 Heiyi Zhuang children aged<16 years old,with 100 cases in asthma group and 100 cases in healthy group.The polymorphisms of the rs1042713 of ADRB2 gene and the rs1801275 of IL-4R gene were detected by PCR-LCR chain reaction,and detection results were analyzed.Results(1)AA,AG and GG genotypes were detected at the rs1042713 of ADRB2 gene and the rs1801275 of IL-4R gene loci.(2)There was statistically significant difference in the genotypes of the rs1042713 of ADRB2 gene and the rs1801275 of IL-4R gene loci between the asthma group and the healthy group(P<0.05).(3)Genotype AA,AG+AA and allele A of rs1042713 of ADRB2 gene might increase the risk of asthma,while GG+AG might decrease the risk of asthma.AG,AG+GG and allele G at the rs1801275 of IL-4R gene could reduce the risk of asthma,while AA+GG could increase the risk of asthma compared with heterozygous AG.Conclusion(1)There are polymorphisms of the rs1042713 of ADRB2 gene and the rs1801275 of IL-4R gene of Heiyi Zhuang children in Guangxi;(2)The rs1042713 of ADRB2 gene and the rs1801275 of IL-4R gene loci may be related factors in the asthma attack of Heiyi Zhuang children in Guangxi.
