摘 要:作为植物的转录因子家族之一,AP2/ERF基因家族在植物的发育调控、激素响应以及调控植物应对各种胁迫的生命过程中发挥关键作用。本研究基于菊花脑(Chrysanthemum nankingense)基因组,筛选到菊花AP2/ERF基因家族,并对该家族的成员进行基因与蛋白结构、蛋白理化性质、保守基序、启动子顺式元件、组织和胁迫条件下的表达特征等进行了相关分析。此外,结合现有的公共数据库的60个菊花转录组测序数据,通过WGCNA分析对该基因家族成员进行了初步的功能分类。研究结果表明:菊花AP2/ERF基因家族共筛选229个成员,分别将其命名为CnAP2001~CnAP2229。系统进化分析发现菊花AP2/ERF家族成员属于4个亚家族(AP2,ERF,DREB,RAV)以及3个Soloist(未分类)序列。通过WGCNA的方法,将筛选的39508个表达的基因划分为36个模块。分析发现有31个AP2/ERF家族成员在花发育相关的7个模块,20个成员在胁迫相关的3个模块。综上所述,AP2/ERF家族成员在菊花花发育过程以及非生物胁迫过程中发挥着重要功能,本研究为菊花分子育种提供了新的思路。As one of the transcription factor families of plants,the AP2/ERF gene family plays a key role in plant developmental regulation,hormone response,and regulation of the life process of plants in response to various stresses.We identified the AP2/ERF gene family from the Chrysanthemum nankingense genome.Gene structure,protein physicochemical properties,conservative motifs,promoter cis-elements,and tissue expressions profiles under stress conditions were analyzed.In addition,combined with 60 transcriptome sequencing data from existing public databases,WGCNA analysis was used to provide a preliminary functional classification of the gene family members.The results showed that a total of 229 members of the Chrysanthemum AP2/ERF gene family were identified.We named them CnAP2001~CnAP2229.Phylogenetic analysis revealed that the members of the Chrysanthemum AP2/ERF family belong to 4 subfamilies(AP2,ERF,DREB,RAV)and 3 unclassified sequences(Soloist).We screened 39508 genes byWGCNA and divided them into 36 modules.There were 31 AP2/ERF family members in 7 modules regulating flower development,and 20 members in 3 modules responding to stresses.In conclusion,the AP2/ERF family members play an important role in flower development and response to abiotic stress in the chrysanthemum.This study provides a new idea for the molecular breeding of Chrysanthemum.
