摘  要:肾纤维化是所有慢性肾脏病的最终通路,转化生长因子β1(TGF-β1)表达增加对其发生发展及预后都有很大影响,而原癌蛋白Ski家族的Ski/SnoN就是这一信号通路的关键负调控因子。本文就TGF-β1通路负调节因子的缺失如何加剧肾损害的形成进行了综述,并讨论了恢复Ski/SnoN的表达在改善肾纤维化中的治疗价值,提出了抑制慢性肾脏病TGF-β1过度激活的新方法,并为今后肾纤维化的诊断、治疗提供参考。Renal fibrosis is the final pathway in all chronic kidney diseases.Elevated expression of transforming growth factorβ1(TGF-β1)is indispensable for its development and prognosis.Ski/SnoN of the proto-oncoprotein Ski family is a key negative regulator of this signaling pathway.This review addresses how the loss of these negative regulators of the TGF-β1 pathway exacerbates the renal lesion formation and discusses the therapeutic value of restoring the expression of Ski/SnoN in ameliorating fibrosis.Thus,a new method to inhibit TGF-β1 over activation in chronic kidney is proposed and it provides a reference for future diagnosis and treatment of renal fibrosis.
