摘 要:目的探讨不同浓度葡萄糖对大鼠胰岛INS-1细胞胰岛素释放量的影响。方法研究时间为2020年9月—2021年6月,将大鼠INS-1胰岛细胞分别培养在正常1640培养基和葡萄糖浓度分别为5.5、16.7、25、33、50 mmol/L的培养液中培养48 h,CCK8检测细胞活力、大鼠胰岛素检测试剂盒检测胰岛素释放量和DCFH-DA探针检测活性氧(ROS)的水平。结果不同浓度葡萄糖的细胞活力比较,差异无统计学意义(P>0.05),但是与对照组(1.047±0.101)比较,糖浓度为50 mmol/L时细胞活力下降(0.727±0.096),差异有统计学意义(P<0.05);随着糖浓度的增加,胰岛素释放量、胰岛素细胞内含量和胰岛素总量较对照组显著下降,糖浓度33 mmol/L时(0.207±0.008)、(0.224±0.014)、(0.451±0.079)ng/mL最低,糖浓度50 mmol/L时(0.315±0.011)、(0.374±0.011)、(0.660±0.109)ng/mL有升高趋势;葡萄糖浓度超过16.7 mM后,细胞内的活性氧水平逐渐升高,均高于对照组,差异有统计学意义(P<0.05)。结论不同浓度葡萄糖对大鼠胰岛INS-1细胞胰岛素释放量的降低程度不同,为高糖损伤模型的建立和寻找具有保护胰岛细胞功能的临床药物提供实验数据。Objective To investigate the effects of different concentrations of glucose on insulin release in rat islet INS-1 cells.Methods From September 2020 to June 2021,cultured rat INS-1 islet cells in normal 1640 medium and culture medium with glucose concentrations of 5.5,16.7,25,33,and 50 mmol/L for 48 hours.CCK8 detected cell viability,rat insulin detection kit detected insulin release and DCFH-DA probe detected the level of reactive oxygen species(ROS).Results There was no statistically significant difference in cell viability among groups with different concentrations of glucose(P>0.05),but compared with the control group(1.047±0.101),the cell viability decreased when the glucose concentration was 50 mmol/L(0.727±0.096),and the difference was statistically significant(P<0.05).Insulin release,intracellular insulin content,and total insulin mass decreased significantly with increasing glucose concentration.As the sugar concentration increased,the insulin release,intracellular insulin content,and total insulin content significantly decreased compared to the control group,and the difference was statistically significant(P<0.05).At a sugar concentration of 33 mmol/L,(0.207±0.008)ng/mL,(0.224±0.014)ng/mL,and(0.451±0.079)ng/mL were the lowest,while at a sugar concentration of 50 mmol/L,(0.315±0.011)ng/mL,(0.374±0.011)ng/mL,and(0.660±0.109)ng/mL showed an upward trend.After the glucose concentration exceeded 16.7 mM,the intracellular reactive oxygen species levels gradually increased,both higher than the control group,the difference was statistically significant(P<0.05).Conclusion Different concentrations of glucose can reduce the insulin release of rat islet INS-1 cells to different degrees,which provides experimental data for the establishment of high glucose injury model and the search for clinical drugs that can protect islet cell function.
