摘 要:目的探讨低氧环境下间充质干细胞在SDF-1复合PLGA/胶原三维支架上的增殖和迁移的影响,为治疗肌腱损伤提供理论依据。方法于2021年6月—2022年6月齐齐哈尔医学院分子生物学实验室实施,购买大鼠骨髓间充质干细胞,随机分为4组,分别为低氧对照组,低氧P/C支架组,低氧S-P/C支架组,正常对照组,每组各进行5次CCK-8细胞增殖实验检测细胞的增殖情况,每组各进行3次Transwell细胞迁移实验检测细胞的迁移情况。结果在第7天各组细胞培养活性显示,低氧对照组吸光度值(1.26±0.10)高于正常对照组的(1.07±0.12),差异有统计学意义(t=2.724,P=0.027)。低于低氧P/C支架组吸光度值(1.50±0.15)高于低氧对照组的(1.26±0.10),差异有统计学意义(t=2.909,P=0.020)。低氧S-P/C支架组吸光度值(1.71±0.13)高于低氧P/C支架组的(1.50±0.15),差异有统计学意义(t=2.486,P=0.038)。各组细胞迁移数量,低氧对照组细胞迁移数量为(35.2±5.4)×10^(3)个相对正常对照组的(21.6±3.5)×10^(3)个,差异有统计学意义(t=3.661,P=0.022)。低氧P/C支架组细胞迁移数量为(47.8±4.9)×10^(3)个高于低氧对照组的(35.2±5.4)×10^(3)个,差异有统计学意义(t=2.993,P=0.040)。低氧S-P/C支架组细胞迁移数量为(56.6±2.3)×10^(3)个高于低氧P/C支架组的(47.8±4.9)×10^(3)个,差异有统计学意义(t=2.816,P=0.048)。结论在低氧环境下,SDF-1复合PLGA/胶原三维支架可促进间充质干细胞的增殖和迁移。Objective To investigate the effect of mesenchymal stem cells on the proliferation and migration of SDF-1 composite PLGA/collagen three-dimensional scaffold under hypoxic environment,so as to provide a theoretical basis for the treatment of tendon injury.Methods From June 2021 to June 2022,the Molecular Biology Laboratory of Qiqihar Medical University purchased rat bone marrow mesenchymal stem cells and randomly divided them into 4 groups:hypoxic control group,hypoxic P/C scaffold group,hypoxic S-P/C scaffold group and normal control group.CCK-8 cell proliferation experiment was performed 5 times in each group to test cell proliferation.Transwell cell migration was performed 3 times in each group to detect the cell migration.Results On the 7th day,the absorbance of hypoxic control group(1.26±0.10)was higher than that of normal control group(1.07±0.12),and the difference was statistically significant(t=2.724,P=0.027).The absorbance of the hypoxic P/C stent group(1.50±0.15)was higher than that of the hypoxic control group(1.26±0.10),and the difference was statistically significant(t=2.909,P=0.020).The absorbance of the hypoxic S-P/C stent group(1.71±0.15)was higher than that of the hypoxic P/C stent group(1.50±0.15),and the difference was statistically significant(t=2.486,P=0.038).The number of cell migration in each group was(35.2±5.4)×10^(3) in the hypoxia control group,compared with(21.6±3.5)×10^(3) in the normal control group,and the difference was statistically significant(t=3.661,P=0.022).The number of cell migration in the hypoxic P/C scaffold group was(47.8±4.9)×10^(3),compared with that in the hypoxic control group(35.2±5.4)×10^(3),and the difference was statistically signifi-cant(t=2.993,P=0.040).The number of cell migration in the hypoxic S-P/C scaffold group was(56.6±2.3)×10^(3),com-pared with(47.8±4.9)×10^(3) in the hypoxic P/C scaffold group,and the difference was statistically significant(t=2.816,P=0.048).Conclusion SDF-1 combined with PLGA/collagen three-dimensional scaffolds can p
