五味子乙素对慢性铝中毒大鼠海马神经细胞凋亡相关基因的影响

摘  要:目的研究五味子乙素(Schisandrin B,Sch B)对慢性铝中毒大鼠海马神经细胞凋亡相关基因的影响。方法用AlCl 3水溶液拌入饲料中连续喂养大鼠建立慢性铝中毒模型,将建模成功的大鼠按随机方法分为5组:铝中毒模型组、阳性对照组和Sch B低剂量组、Sch B中剂量组、Sch B高剂量组3个剂量实验组。阳性对照组用吡拉西坦按40 mg/kg剂量灌胃给药,Sch B低剂量组、Sch B中剂量组、Sch B高剂量组用Sch B分别按10mg/kg、20 mg/kg、40 mg/kg的剂量进行灌胃,另设空白对照组,每组5只。RT-PCR检测大鼠海马神经细胞Caspase-3、Bcl-2、Bax mRNA表达情况。结果RT-PCR结果显示,与空白对照组相比,铝中毒模型组Caspase-3和Bax mRNA表达均明显增多,Bcl-2 mRNA表达明显减少,差异均有统计学意义(P<0.01);与铝中毒模型组相比,Sch B低剂量组、Sch B中剂量组、Sch B高剂量组Caspase-3 mRNA相对表达量明显降低,差异均有统计学意义(P<0.01);Sch B中剂量组、Sch B高剂量组的Bcl-2 mRNA表达升高,差异有统计学意义(P<0.05或P<0.01);Sch B中剂量组、Sch B高剂量组的Bax mRNA表达降低,差异有统计学意义(P<0.05或P<0.01)。结论Sch B可以通过对Caspase-3、Bcl-2、Bax mRNA表达调控,影响海马神经细胞的凋亡。Objective To investigate the impact of Schisandrin B(Sch B)on apoptosis-related genes in hippocampal neurons of rats with chronic aluminum toxicity.Methods A chronic aluminum toxicity model was established by continuously feeding rats with AlCl 3 aqueous solution mixed in their feed.The successfully modeled rats were randomly divided into five groups:the aluminum toxicity model group,positive control group,and low,medium,and high-dose Sch B experimental groups.The positive control group received intragastric administration of piracetam at a dose of 40 mg/kg,while the Sch B low,medium,and high-dose groups received intragastric administration of Sch B at doses of 10 mg/kg,20 mg/kg,and 40 mg/kg,respectively.A blank control group was also included,with five rats in each group.The expression of Caspase-3,Bcl-2,and Bax mRNA in hippocampal neurons of rats was measured using RT-PCR.Results RT-PCR results showed that compared with the blank control group,the expression of Caspase-3 and Bax mRNA was significantly increased in the aluminum toxicity model group,while the expression of Bcl-2 mRNA was significantly decreased,with statistically significant differences(P<0.01).When compared to the aluminum toxicity model group,the Sch B low,medium,and high-dose groups showed significantly decreased relative expressions of Caspase-3 mRNA,with statistically significant differences(P<0.01).The expression of Bcl-2 mRNA was increased in the Sch B medium and high-dose groups,with statistically significant differences(P<0.05 or P<0.01).The expression of Bax mRNA was decreased in the Sch B medium and high-dose groups,with statistically significant differences(P<0.05 or P<0.01).Conclusion Schisandrin B can affect the apoptosis of hippocampal neurons by regulating the expression of Caspase-3,Bcl-2,and Bax mRNA.
