摘 要:目的:研究栀子提取物对抑郁症的治疗作用及机制。方法:将60只小鼠随机分为空白组、模型组、氟西汀组、栀子总提物组、栀子苷组,除空白组外其他各组均采用慢性不可预知应激(CUMS)构建抑郁模型。造模后,氟西汀组给予氟西汀20 mg/kg,栀子总提物组给予栀子总提物组(相当于原味药)6 g/kg,栀子苷组给予栀子苷100 mg/kg,空白组和模型组给予生理盐水10 mL/kg,各组连续治疗3周。观察各组小鼠体重变化情况、糖水偏好实验、旷场实验(活动总距离和中央活动时间)、悬尾实验与强迫游泳实验不动时间、皮质酮(CORT)、促肾上腺皮质激素(ACTH)、5-羟色胺(5-HT)、脑源性神经营养因子(BDNF)、糖皮质激素受体(GR)、盐皮质激素受体(MR)水平及MR、GR、BDNF蛋白表达情况。结果:(1)在造模4周时模型组、栀子总提物组、栀子苷组、氟西汀组体重开始显著下降,至第7周时各造模组小鼠体重与空白组比较,差异有统计学意义(P<0.05)。(2)与空白组相比,模型组小鼠糖水偏好率显著下降,中央活动时间、强迫游泳和悬尾不动时间显著增加,差异有统计学意义(P<0.001)。与模型组比较,栀子总提物组、栀子苷组、氟西汀组糖水偏好率显著升高,栀子总提物组、栀子苷组、氟西汀组小鼠中央活动时间、强迫游泳和悬尾不动时间显著降低,差异有统计学意义(P<0.05)。各组小鼠自主活动总距离比较差异无统计学意义(P>0.05)。(3)与空白组相比,模型组小鼠CORT、ACTH、MR水平显著升高,5-HT、BDNF、GR水平显著降低,差异有统计学意义(P<0.05)。与模型组相比,栀子总提物组、栀子苷组、氟西汀组小鼠CORT、ACTH、MR水平显著降低,5-HT、BDNF、GR水平显著升高,差异有统计学意义(P<0.05)。结论:栀子提取物具有显著的抗抑郁作用,其作用机制可能和调节下丘脑-垂体-肾上腺(HPA)轴功能,下调CORT、ACTH、MR水平及升高BDNF、5-HT、GR的水Objective:To study the therapeutic effect and mechanism of Gardenia jasminoides Ellis extract for depression.Methods:Sixty mice were randomly divided into blank group,model group,fluoxetine group,Gardenia jasminoides Ellis total extract group,and geniposide group.Except the blank group,the remaining groups used CUMS to construct depression models.After successful modeling,fluoxetine group was given 20 mg/kg fluoxetine,Gardenia jasminoides Ellis total extract group was given 6 g/kg Gardenia jasminoides Ellis total extract(equivalent to the original drug),geniposide group was given 100 mg/kg geniposide,blank group and model group were given 10 mL/kg physiological saline.Each group was treated continuously for 3 weeks.The changes of mice body weight,sucrose preference test,open field test(total activity distance and central activity time),tail suspension test and forced swim test immobility time,cortisol(CORT),adrenocorticotropic hormone(ACTH),serotonin(5-HT),brain-derived neurotrophic factor(BDNF),glucocorticoid receptor(GR),mineralocorticoid receptor(MR)level,and MR,GR and BDNF protein expression levels were observed in each group.Results:①At 4 weeks of modeling,the body weight in the model group,Gardenia jasminoides Ellis total extract group,geniposide group and fluoxetine group began to decrease significantly,and there were statistically significant differences in the mice body weight between each modeling group and blank group at 7 weeks(P<0.05).②Compared with the blank group,the sucrose preference rate of mice decreased significantly,while the central activity time,forced swim and tail suspension immobility time increased significantly in the model group,and the difference was statistically significant(P<0.001).Compared with the model group,the sucrose preference rate increased significantly in the Gardenia jasminoides Ellis total extract group,geniposide group and fluoxetine group,while the central activity time,forced swim and tail suspension immobility time decreased significantly in the Gardenia jasmin
