摘 要:目的探讨硫酸卡那霉素慢性损害后豚鼠听皮层蛋白质组的变化,以及电针对抑制素和吞蛋白-A1表达的影响。方法105只成年健康杂色豚鼠随机分为对照组(7只)、硫酸卡那霉素模型组(49只,模型组)、硫酸卡那霉素+电针组(49只,电针组)。模型组与电针组连续注射硫酸卡那霉素7天,电针组注射半小时后予以电针翳风穴和听宫穴15分钟,连续7天,对照组注射等量生理盐水;模型组与电针组根据药物注射后时间再分为注射后1、7、14、28、56、70、140天组(每组7只)。采用双向电泳技术、质谱鉴定分析筛选并切取21个表达差异明显的蛋白点进行鉴定,蛋白质印迹法检测抑制素与吞蛋白-A1的表达。结果①灰度差异2倍以上的差异表达蛋白质有抑制素、吞蛋白-A1等18种。②与对照组相比,模型组注射后第14、28、56天抑制素表达降低(P<0.05),第1、14天吞蛋白-A1表达增高(P<0.05);与模型组相比,电针组注射后第7、70天抑制素表达增高(P<0.05),第14、28、56天吞蛋白-A1表达降低(P<0.05)。结论抑制素等18种蛋白质可能参与了硫酸卡那霉素致豚鼠听皮层慢性损害的过程,电针听宫穴与翳风穴可能通过调节抑制素、吞蛋白-A1的表达促进听皮层的重塑进程。Objective To study the changes of the proteome in auditory cortex after chronic kanamycin-induced damage,and the effects of electroacupuncture on the expression of Prohibition and Endophilin-A1.Methods A total of 105 healthy adult variegated guinea pigs were randomly divided into three groups,including control group(CG,n=7)kanamycin treatment group(KTG,n=49)and kanamycin sulfate with electroacupuncture treatment group(KEG,n=49).KTG and KEG were injected with kanamycin sulfate for 7 days,while KEG were electroacupunctured at Yifeng and Tinggong for 15 minutes after injected the kanamycin for half an hour.The control group(CG,n=7)was injected with the same dose of normal saline.KTG and KEG were divided into 7 groups(n=7)for 1,7,14,28,56,70 and 140 days after injection.Expression differences of 21 protein spots were screened and identified by two-dimensional electrophoresis and mass spectrometry.The expressions of Prohibition and Endophilin-A1 were detected by Western blot.Results①There were 18 differentially expressed proteins with more than 2-fold gray difference,such as Prohibition and Endophilin-A1.②Compared with the CG,the expression of Prohibition in KTG declined on the 14th,28th and 56th days after injection(P<0.05),and the expression of Endophilin-A1 in KTG increased on the 1st and 14th days after injection(P<0.05).Compared with the KTG,the expression of Prohibition in KEG increased on the 7th and 70th days after injection(P<0.05),and Endophilin-A1 in KEG declined on the 14th,28th and 56th days after injection(P<0.05).Conclusion Eighteen proteins such as Prohibition may be involved in the process of chronic damage caused by kanamycin sulfate.Electroacupuncture at Tinggong and Yifeng points may enhance the remodeling process of auditory cortex by regulating the expression of Prohibition and Endophilin-A1.
