摘 要:目的探讨2型糖尿病(T2DM)大鼠血清中蛋白结合态铜(Cu)、锌(Zn)的水平。方法30只成年Sprague Dawley(SD)大鼠随机分为对照组(n=10,普通饲料)和T2DM模型组[n=20,高糖高脂饲料,腹腔注射链脲佐菌素(STZ)诱导造模],分别取2组大鼠心脏血离心检测血清中的Cu、Zn元素总形态及非蛋白结合形态;于0~300μg/L制作Cu、Zn元素的标准曲线,获取检定限和定量限;通过精密度实验,计算Cu、Zn总含量、非蛋白结合态含量及相对标准偏差(RSD);采用连续光源石墨炉原子吸收光谱法检测血清Cu、Zn总形态含量和非蛋白结合态含量,计算蛋白结合态含量。结果Cu、Zn元素在标准曲线范围内的相关系数(r)>0.999,RSD均<5%,加标回收率为87%~110%;与对照组相比,T2DM模型组大鼠血清中Cu总形态含量和蛋白结合态含量均升高(P<0.05),Zn总形态含量和蛋白结合态含量均降低(P<0.05),Cu、Zn总形态含量和蛋白结合态元素含量的比值均升高(P<0.05);大鼠血清Cu总形态含量和蛋白结合态含量与Zn总形态含量和蛋白结合态含量呈负相关关系(P<0.05)。结论T2DM大鼠血清中Cu、Zn总形态变化主要由蛋白结合态变化引起,测定2种元素的蛋白结合态含量较总形态含量可以更好地反映它们在DM过程中的生物学作用。Objective To investigate the levels of protein-bound copper(Cu)and zinc(Zn)elements in serum of rats with type 2 diabetes mellitus(T2DM).Methods Thirty adult Sprague Dawley(SD)rats were randomly divided into control group(n=10,normal diet)and T2DM model group[n=20,high glucose and high fat diet,induced by peritoneal injection of streptozotocin(STZ)].The total morphology of Cu and Zn elements and non-protein binding morphology in the serum were detected by centrifugation in the heart blood of rats of 2 groups.The standard curves of Cu and Zn elements were prepared from 0-300μg/L,and the test and quantification limit were obtained.The total Cu,Zn content,non-protein bound state content and relative standard deviation(RSD)were calculated by precision experiments.Serum Cu and Zn total morphological content and non-protein bound state were measured by continuous light source,and the content of protein-bound state was calculated.Results The correlation coefficient(r)of Cu and Zn elements within the standard curve range were>0.999,and all the RSD were<5%.The spike recovery ranged from 87%to 110%.The recovery rate of the spike was 87%-110%.Compared with the control group,the total Cu and protein bound state in the serum of T2DM model group was increased(P<0.05),and the total Zn and protein bound state was decreased(P<0.05).The ratio of the total morphological content and the element content of Cu and Zn in the protein-bound state were all increased(P<0.05).Rat serum Cu total morphology content and protein bound state content as well as Zn total morphology content and protein bound state content were inversely correlated(P<0.05).Conclusion The total morphological changes of Cu and Zn in the serum of T2DM rats are mainly caused by the changes of protein-binding state.Testing the protein-binding state content of the two elements can better reflect their biological role in DM than the total morphological content.
