摘 要:本研究基于网络药理学预测大黄发挥清热解毒作用的质量标志物。利用TCMSP数据库、Swiss Target Prediction服务器等筛选大黄的主要活性成分并预测其发挥清热解毒潜在作用靶点及其通路。结果表明:大黄主要通过泽兰黄醇、番泻苷、大黄酸、β-谷甾醇等14种化合物,作用于287个靶点,其中SRC、TP53、JUN、HSP90AA1、ESR1为核心靶点;主要通过钙离子、花生四烯酸代谢、TRP通道炎性介质调节等信号通路发挥清热解毒作用。由此预测到大黄发挥清热解毒作用的质量标志物为番泻苷、大黄酸,为其作用机制的进一步研究奠定了基础。Based on the theory and method of network pharmacology,this study predicted the quality markers of rhubarb's clearing away heat and detoxification effects.The TCMSP platform was used to screen the main active components of rhubarb and predict its potential targets.The results showed that rhubarb mainly acted on 287 targets through 14 compounds,such as eupatorium adenophorum,sennoside,rhein andβ-sitosterol,among which the key targets were SRC,TP53,JUN,HSP90AA1 and ESR1.The heat-clearing and detoxification effects of rhubarb were mainly mediated by signal pathways such as calcium,arachidonic acid metabolism,and regulation of inflammatory mediators in TRP channels.In this experiment,the quality markers of rhubarb's heat-clearing and detoxicating effects predicted sennoside and rhein,which laid a foundation for further research on the mechanism of rhubarb's heat-clearing and detoxicating effects.
