摘  要:以大球盖菇菌丝体为试材,在单因素试验基础上,以原生质体产量为响应值,采用响应面方法优化其制备条件,研究了大球盖菇原生质体制备最佳条件,以期为后期大球盖菇菌种遗传改良提供参考依据。结果表明:经响应面试验优化后,获得大球盖菇原生质体的最佳制备条件为酶解时间2.5 h、酶解温度32.2℃、菌龄3.8 d,此时获得大球盖菇原生质体产量最大,为35.8×10^(6)CFU·mL^(-1),与理论产量(35.15×10^(6)CFU·mL^(-1))偏差较小。因此,基于响应面法确定的最佳制备条件可获得较高的原生质体产量。The mycelium of Stropharia rugosoannulate was used as the research material.The reaction conditions were optimized by response surface method based on the single factor test with protoplast yield as response value,in order to provide reference for genetic improvement of S.rugosoannulatain the later stage.The results showed that after the optimization of response surface method,the preparation conditions for the protoplasts were obtained as follows,enzymolysis time was 2.5hours,enzymolysis temperature was 32.2℃,and the mycelial age was 3.8days,the maximum protoplast yield was 35.8×10^(6) CFU·mL^(-1),Compared with the theoretical yield(35.15×10^(6) CFU·mL^(-1)),the deviation was small.Therefor,the optimum preparation conditions based on response surface method can obtain higher protoplast yield.
