摘 要:目的观察优化颠倒散的配方比例及配制方法对痤疮常见致病菌(表皮葡萄球菌、金黄色葡萄球菌、痤疮丙酸杆菌)的体外抑菌效果。方法优化颠倒散配方比例,按大黄∶硫黄比例不同分为9组(1∶0、1∶1、1∶2、1∶3、1∶4、0∶1、2∶1、3∶1和4∶1),检测其分别对痤疮常见致病菌的体外抑菌活性;以配制方法、溶剂类型及溶剂量为影响因素,以抑菌圈直径为评价指标,采用L_(9)(3^(3))正交实验优化颠倒散的调配方法。结果当大黄∶硫黄为2∶1时,对表皮葡萄球菌、痤疮丙酸杆菌平均抑菌圈直径均为(25.50±1.50)mm;对金黄色葡萄球菌当大黄∶硫黄为2∶1及3∶1时抑菌圈直径相同,均为(22.00±1.00)mm。正交设计对表皮葡萄球菌最佳抑菌组合为B_(2)A_(3)C_(3)(研匀+5%乳化剂+25%甘油4 mL),对痤疮丙酸杆菌最佳抑菌组合为A_(3)B_(1)C_(2)(研匀+5%乳化剂+蒸馏水3 mL),金黄色葡萄球菌最佳抑菌组合为C_(3)B_(3)A_(3)(研匀+5%乳化剂+25%乙醇4 mL)。结论通过改变颠倒散的配方比例及优化颠倒散的配制方法,均可增强抑菌作用。Objective In order to observe antibacterial effects in vitro of Dian-dao San using improved formula ratio and different preparation methods against common pathogenic bacteria of acne[Staphylococcus epidermidis(S.epidermidis),Staphylococcus aureus(S.aureus),Propionibacterium acnes(P.acnes)].Methods The optimized formulation(rhubarb and sulphur)was composed of 1∶0,1∶1,1∶2,1∶3,1∶4,0∶1,2∶1,3∶1,4∶1,a total of nine groups,respectively detected their antibacterial activity in vitro.The preparation conditions,including preparation method,solvent type and solvent amount,were optimized by an L_(9)(3^(3))orthogonal experiment.Results When the ratio of rhubarb and sulphur was set as 2∶1,the average diameter of inhibition zone against S.epidermids and P.acnes was(25.50±1.50)mm.For S.aureus,the diameter of the inhibitory zone value were the same when the ration was set as either 2∶1 or 3∶1,both were(22.00±1.00)mm.Under the orthogonal design,the best antibacterial combination for S.epidermids was B_(2)A_(3)C_(3)(grinding well with 5% emulgator and 4 mL 25% glycerol).The best for P.acnes was A_(3)B_(1)C_(2)(grinding well with 5% emulgator and 3 mL distilled water).The best for S.aureus was C_(3)B_(3)A_(3)(grinding well with 5% emulgator and 4 mL 25% ethanol).Conclusion The antibacterial effects can be enhanced by improving formula ratio and optimizing the compound method of Dian-dao San.
