摘 要:目的探讨circ_核糖核蛋白(LARP)4对膀胱癌细胞T24增殖、迁移和凋亡的影响及分子机制。方法选取膀胱癌患者的癌组织及癌旁组织,实时荧光定量聚合酶链反应(RT-qPCR)检测circ_LARP4和miR-135a-5p的表达水平;Western印迹法检测原钙黏蛋白(PCDH)9蛋白表达;将T24细胞随机分为pcDNA-circ_LARP4组、pcDNA组、anti-miR-135a-5p组、anti-miR-NC组、pcDNA-circ_LARP4+miR-135a-5p组;用四甲基偶氮唑蓝(MTT)法检测细胞增殖抑制率;克隆形成实验检测集落形成数;细胞移动范围采用划痕实验进行检测;采用流式细胞术检测细胞凋亡;双荧光素酶报告实验检测circ_LARP4、miR-135a-5p、PCDH9之间的靶向关系。结果与癌旁组织相比,膀胱癌组织中circ_LARP4和PCDH9蛋白的表达水平显著降低,miR-135a-5p表达水平显著升高(P<0.05)。过表达circ_LARP4或抑制miR-135a-5p表达,PCDH9蛋白表达水平升高,miR-135a-5p表达水平降低,T24增殖抑制率升高,集落形成数减少,迁移距离缩短,细胞凋亡率升高,差异有统计学意义(P<0.05)。过表达miR-135a-5p可逆转circ_LARP4对T24细胞增殖、迁移的抑制和凋亡的诱导作用。circ_LARP4靶向miR-135a-5p,miR-135a-5p靶向PCDH9。结论circ_LARP4通过调控miR-135a-5p/PCDH9轴抑制膀胱癌细胞T24的增殖、迁移,诱导细胞凋亡。Objective To explore the effect of circ_ribonucleoprotein(LARP)4 on the proliferation,migration and apoptosis of bladder cancer cell T24 and its molecular mechanism.Methods The cancer tissues and adjacent tissues of bladder cancer patients were selected,and the expression levels of circ_LARP4 and miR-135a-5p were detected by real-time fluorescent quantitative polymerase chain reaction(RT-qPCR);protocadherin(PCDH)9 protein expression was detected by Western blot method;T24 cells were randomly divided into pcDNA-circ_LARP4 group,pcDNA group,anti-miR-135a-5p group,anti-miR-NC group,pcDNA-circ_LARP4+miR-135a-5p group;methylthiazol tetrazolium(MTT)method was used to detect cell proliferation inhibition rate;colony formation test was used to detect colony formation number;scratch test was used to detect cell migration distance;flow cytometry was used to detect cell apoptosis;dual luciferase reporter test was used to detect the targeting relationship of circ_LARP4,miR-135a-5p and PCDH9.Results Compared with adjacent tissues,the expression levels of circ_LARP4 and PCDH9 protein in bladder cancer tissues were significantly decreased,and the expression of miR-135a-5p was significantly increased(P<0.05).Overexpression of circ_LARP4 or inhibition of miR-135a-5p expression,expression level of PCDH9 protein was increased,expression level of miR-135a-5p was decreased,proliferation inhibition rate of T24 was increased,colony formation number was decreased,migration distance was shortened,and cell apoptosis rate was increased,the difference was statistically significant(P<0.05).Overexpression of miR-135a-5p could reverse the inhibition of circ_LARP4 on the proliferation and migration of T24 cells and the induction of apoptosis.circ_LARP4 targeted miR-135a-5p,and miR-135a-5p targeted PCDH9.Conclusions circ_LARP4 could inhibit the proliferation and migration of bladder cancer cell T24 and induce cell apoptosis by regulating the miR-135a-5p/PCDH9 axis.
