摘　　要：目的建立同时测定追风透骨丸中龙胆苦苷和芍药苷含量的高效液相色谱法。方法色谱柱为Agilent Eclipse Plus C18柱(250 mm×4.6 mm,5μm),流动相为甲醇-0.1%磷酸溶液(30∶70,V/V),流速为1.0 mL/min,检测波长为230 nm,柱温为30℃,进样量为10μL。结果龙胆苦苷和芍药苷质量浓度分别在15.718~157.175μg/mL和16.095~160.949μg/mL范围内与峰面积线性关系良好(r=0.9999,n=6);精密度、稳定性、重复性试验结果的RSD均小于2.0%;平均加样回收率分别为99.77%和99.28%,RSD分别为0.66%和0.60%(n=9)。12批样品中龙胆苦苷和芍药苷的平均含量分别为0.856,1.174 mg/g。结论该方法操作简便、结果准确、重复性及专属性良好,可用于同时测定追风透骨丸中龙胆苦苷和芍药苷的含量。Objective To establish a high-performance liquid chromatography(HPLC)method for the simultaneous determination of gentiopicroside and paeoniflorin in Zhuifeng Tougu Pills.Methods The chromatographic column was Agilent Eclipse Plus C18 column(250 mm×4.6 mm,5μm),the mobile phase was methanol-0.1%phosphoric acid solution(30∶70,V/V),the flow rate was 1.0 mL/min,the detection wavelength was 230 nm,the column temperature was 30℃,and the injection volume was 10μL.Results The linear ranges of gentiopicroside and paeoniflorin were 15.718-157.175μg/mL(r=0.9999)and 16.095-160.949μg/mL(r=0.9999,n=6).The RSDs of precision,stability,and repeatability test results were lower than 2.0%.The average recoveries of gentiopicroside and paeoniflorin were 99.77%and 99.28%with RSDs of 0.66%and 0.60%(n=9),respectively.The average contents of gentiopicroside and paeoniflorin in 12 batches of samples were 0.856 mg/g and 1.174 mg/g,respectively.Conclusion This method is simple and accurate with good repeatability and specificity,which can be used for the simulaneous determination of gentiopicroside and paeoniflorin in Zhuifeng Tougu Pills.
