摘  要:目的:通过比较生品与麸炒苍术对脾虚证大鼠小肠组织结构及转运相关蛋白载体的影响,探讨麸炒法影响苍术改善胃肠功能的可能机制。方法:雄性SD大鼠70只随机分为正常组、脾虚证模型组(以下简称模型组)、生苍术高、低剂量组、麸炒苍术高、低剂量组、复方谷氨酰胺组,每组10只。除正常组外的6组均以苦寒破气加饥饱失常法复制脾虚模型,造模时间为21 d。完成造模后,各治疗组每天按相应剂量药物灌胃治疗,共计14 d:生苍术高、低剂量组10、2.5 g·kg^(-1);麸炒苍术高、低剂量组10、2.5 g·kg^(-1),复方谷氨酰胺组9 mg·kg^(-1);正常组、模型组给予同体积生理盐水灌胃。治疗期间行脾虚大鼠一般生存状况、宏观证候评分及每日体质量增加量和摄食量、肛温测定,治疗期满后处死大鼠,苏木素-伊红(HE)染色观察各组大鼠小肠组织病理学改变,酶联免疫吸附测定法(ELISA)检测血清5-羟色胺(5-HT)含量,比色法检测血清D-木糖、乳酸、淀粉酶含量,考马斯亮蓝(Bradford)法检测小肠组织游离脂肪酸受体3(FFA3)、肽转运载体1(PepT1)含量水平,免疫组化法检测小肠组织中钠依赖性葡萄糖转运体1(SGLT1)、葡萄糖转运体1(GLUT1)蛋白表达,实时荧光定量聚合酶链式反应(Real-time PCR)检测葡萄糖转运体2(GLUT2)、钠/氢交换因子3(NHE3)、5-羟色胺受体4(5-HT4R)mRNA表达。结果:与正常组比较,模型组大鼠造模结束时,大部均表现为精神迟钝,眯眼食少,倦怠少动等脾虚证表现,小肠黏膜上皮及固有膜内基本结构被破坏,血清乳酸、5-HT含量升高,血清淀粉酶及D-木糖含量降低(P<0.01),与模型组比较,各个治疗组一般表现不同程度好转,小肠微观结构不同程度修复,体质量每日增加量、肛温及脾虚宏观证候积分不同程度好转(P<0.05,P<0.01),血清乳酸、5-HT含量不同程度降低,血清淀粉酶及D-木糖含量不同程度升高(P<0.05,P<0.01),小肠Objective:To explore the possible mechanism of bran-fried method effecting atractylosis to improve gastrointestinal function by comparing the effects of raw and bran-fried atractylodes on intestinal structure and transport-related protein carriers in rats with spleen deficiency syndrome.Methods:Seventy male SD rats were randomly divided into normal group,spleen deficiency syndrome model group(hereinafter referred to as model group),raw atractylodes(high and low)dose group,bran-fried atractylodes(high and low)dose group and compound glutamine group,10 rats in each group.The six groups except the normal group were used to replicate the spleen deficiency model by the method of bitter cold and broken qi combined with hunger and satiation disorder,and the modeling time was 21 days.After modeling,each treatment group was given the corresponding dose drugs by gavage every day for 14 days:the high and low dose groups of raw atractylodes were 10,2.5 g•kg^(-1);bran-fried atractylodes high and low dose groups were 10,2.5 g•kg^(-1),and compound glutamine group was 9 mg•kg^(-1);Normal group and model group were given the same volume of normal saline During the treatment,the general survival status,macroscopic syndrome score,daily weight gain,food intake and anal temperature of the spleen-deficient rats were measured.After the treatment,the rats were killed.Hematoxylin eosin(HE)staining was performed to observe the histopathological changes of the small intestine of the rats,and serum 5-hydroxytryptamine(5-HT)content was detected by enzyme-linked immunoadsorption assay(ELISA).Serum D-xyloss,lactic acid and amylase contents were determined by colorimetric method.Free fatty acid receptor 3(FFA3)and peptide transporter 1(PepT1)contents in small intestine were determined by Bradford method.The protein expressions of sodium-dependent glucose transporter 1(SGLT1)and glucose transporter 1(GLUT1)were detected by immunohistochemistry.The gene expressions of glucose transporter 2(GLUT2),sodium/hydrogen exchange factor 3(NHE3)and 5
