汉黄芩素对气虚模型LLC种植瘤小鼠的抗肿瘤作用及淋巴细胞亚群的影响

摘　　要：目的:探讨汉黄芩素对气虚模型LLC种植瘤小鼠的抗肿瘤作用及淋巴细胞亚群的影响。方法:将30只SPF级C57BL/6雄性小鼠(N型)用烟熏法建立小鼠气虚模型后,构建气虚型LLC肺癌皮下种植瘤模型。将造模成功的24只小鼠随机分为气虚荷瘤组、汉黄芩素高剂量组、汉黄芩素中剂量组、汉黄芩素低剂量组,每组6只。另取6只正常小鼠作为正常组。汉黄芩素低、中、高剂量组小鼠灌胃给予相应剂量的汉黄芩素混悬液;正常组、气虚荷瘤组小鼠灌胃给予等体积生理盐水,1次/d,连续给药15 d。给药过程中每4 d测量1次体质量和肿瘤体积。给药结束后剖取小鼠肿瘤、脾脏和胸腺称定质量,并采用HE染色法对肿瘤、肝脏和肾脏进行组织病理学检查;采用流式细胞术分析小鼠外周血中的辅助T细胞(CD3+、CD4+)、杀伤T细胞(CD3+、CD8+)、调节性T细胞(Treg)(CD3+、CD4+、CD25+、Foxp3+)、NK细胞(CD3-、CD19+)和B细胞(CD3-、CD49b+)的比例,并计算CD4+/CD8+比值。结果:与正常组比较,气虚荷瘤组小鼠喜蜷卧,活动度减弱,毛发脱落且蓬松凌乱,毛色发灰,欠光泽,精神萎靡,饮食量较正常组小鼠下降。给药结束后,5组小鼠去瘤体质量比较,差异无统计学意义(P>0.05)。与气虚荷瘤组比较,汉黄芩素低、中、高剂量组小鼠瘤体体积和瘤体质量均明显降低(P<0.05或P<0.01),抑瘤率分别为55.68%、56.84%、76.33%。与正常组比较,气虚荷瘤组小鼠脾脏指数明显升高(P<0.05),胸腺指数明显降低(P<0.05);汉黄芩素低、中、高剂量组小鼠脾脏指数、胸腺指数与气虚荷瘤组比较,差异无统计学意义(P>0.05)。HE染色显示,与气虚荷瘤组比较,汉黄芩素低、中、高剂量组小鼠瘤体组织可见较多坏死区域,且肿瘤细胞的排列相对紊乱;肝肾组织均未见明显病变。与正常组比较,气虚荷瘤组小鼠外周血CD3+CD4+T细胞比例、CD3+CD8+T细胞比例、CD4+/CD8+比值、CD3-CD19+B细胞�Objective:To investigate the effect of wogonin on the antitumor effect and lymphocyte subsets of Qi-deficiency model LLC implanted tumor mice.Method:30 SPF grade C57BL/6 male mice(N-type)were used to establish a mouse model of Qi deficiency using smoking method.And then a subcutaneous implanted tumor model of Qi deficiency type LLC lung cancer was constructed.24 mice that successfully established the model were randomly divided into Qi deficiency tumor-bearing group,wogonin high-dose,medium-dose,and low-dose group,with 6 mice in each group.Take another 6 normal mice as the normal group.Mice in low,medium and high dose groups of wogonin were given corresponding doses of wogonin suspension by intragastriction.Mice in the normal group and the Qi deficiency tumor-bearing group were given equal volume of normal saline by instillation once a day for 15 days.The body mass and tumor volume were measured every 4 days during the administration.After the administration,the tumors,spleen and thymus of mice were dissected and weighed,and the tumors,liver and kidney were histopathologically examined by HE staining.The proportions of helper T cells(CD3^(+),CD4^(+)),killer T cells(CD3^(+),CD8^(+)),regulatory T cells(Treg)(CD3^(+),CD4^(+),CD25^(+),Foxp3^(+)),NK cells(CD3-,CD19^(+))and B cells(CD3-,CD49b^(+))in peripheral blood of mice were analyzed by flow cytometry,and the ratio of CD4^(+)/CD8^(+)T cells was calculated.Results:Compared with the normal group,the mice in the Qi deficiency tumor-bearing group liked to curl up,the mobility was weakened,the hair fell off and was fluffy and messy,the hair color was gray,less shiny,and the spirit was listless,and the amount of diet was decreased compared with the normal group mice.After the administration,there was no significant difference in the tumor removal mass among the five groups(P>0.05).Compared with the Qi deficiency tumor-bearing group,the tumor volume and weight of mice in the low,medium,and high dose groups of wogonin were significantly reduced(P<0.05 or P<0.01),and the tu
