摘 要:目的:优化玄参种芽提取工艺。方法:建立玄参种芽中哈巴苷、哈巴俄苷含量测定和方法,采用单因素试验,考察甲醇浓度、料液比和浸泡时间3个因素对哈巴苷、哈巴俄苷含量及总含量的影响。结合Box-Behnken响应面法,以哈巴苷、哈巴俄苷总含量为响应指标,考察甲醇浓度、料液比和浸泡时间3个因素对提取工艺的影响。结果:哈巴苷、哈巴俄苷分别在15.63~500.00μg/mL、10.63~340.00μg/mL范围内线性关系良好,哈巴苷、哈巴俄苷平均回收率分别为100.24%、100.56%,RSD分别为1.98%、1.19%。根据回归方程,结合实际操作确定的最佳提取工艺为甲醇浓度44%、料液比1∶51、浸泡25 min,理论总含量为1.84%。结论:甲醇浓度、料液比和浸泡时间对玄参种芽提取有显著影响,得到的工艺稳定可重复,与理论值接近,可为玄参种芽提取技术规范与标准提供理论基础。Objective:To optimize the extraction process of Xuan Shen(Scrophulariae Radix)seed buds.Method:To establish a method for determining the content of harpagide and harpagoside in the seed buds of Xuan Shen(Scrophulariae Radix).A single factor experiment was conducted to investigate the effects of three factors:methanol concentration,solid-liquid ratio,and soaking time on the content and total content of harpagide and harpagoside.Combined with Box-Behnken response surface methodology,the total content of harpagide and harpagoside were used as response indicators to investigate the effects of methanol concentration,solid-liquid ratio,and soaking time on the extraction process.Results:The linear relationship between harpagide and harpagoside was good within the range of 15.63-500.00μg/mL and 10.63-340.00μg/mL,respectively.The average recovery rates of harpagide and harpagoside were 100.24%and 100.56%,with RSDs of 1.98%and 1.19%,respectively.According to the regression equation,combined with the practical operation,the optimum extraction process was determined as follows:methanol concentration 44%,material liquid ratio 1:51,soaking time 25 min,and theoretical total content 1.84%.Conclusion:The concentration of methanol,the ratio of material to liquid and the soaking time have significant effects on the extraction of the seed buds of Xuan Shen(Scrophulariae Radix).The obtained process is stable and repeatable,close to the theoretical value.It provides a theoretical basis for the technical specifications and standards of seed bud extraction of Xuan Shen(Scrophulariae Radix).
